Project description:Controlled hypobaria presents biology with an environment that is never encountered in terrestrial ecology, yet the apparent components of hypobaria are stresses typical of terrestrial ecosystems. High altitude, for example, presents terrestrial hypobaria always with hypoxia as a component stress, since the relative partial pressure of O2 is constant in the atmosphere. Laboratory-controlled hypobaria, however, allows the dissection of pressure effects away from the effects typically associated with altitude, in particular hypoxia, as the partial pressure of O2 can be varied. In this study, whole transcriptomes of plants grown in ambient (97 kPa/pO2 = 21 kPa) atmospheric conditions were compared to those of plants transferred to five different atmospheres of varying pressure and oxygen composition for 24 h: 50 kPa/pO2 = 10 kPa, 25 kPa/pO2 = 5 kPa, 50 kPa/pO2 = 21 kPa, 25 kPa/pO2 = 21 kPa, or 97 kPa/pO2 = 5 kPa. The plants exposed to these environments were 10 day old Arabidopsis seedlings grown vertically on hydrated nutrient plates. In addition, 5 day old plants were also exposed for 24 h to the 50 kPa and ambient environments to evaluate age-dependent responses. The gene expression profiles from roots and shoots showed that the hypobaric response contained more complex gene regulation than simple hypoxia, and that adding back oxygen to normoxic conditions did not completely alleviate gene expression changes in hypobaric responses.

Project description:Council Regulation (EC) No 1099/2009 on the protection of animals at the time of killing lists in Annex I the stunning interventions currently allowed in the EU, together with the related conditions under which those interventions can be implemented. The regulation allows the Commission to amend Annex I, listing additional stunning interventions, provided they ensure a level of animal welare at least equivalent to that ensured by the one already approved. EFSA was requested to perform such assessment with regard to the implementation of the low atmospheric pressure stunning (LAPS) system on broiler chickens. The ad hoc Working Group (WG) set up by EFSA performed the assessment in three main steps, i.e. checking the data provided against the criteria laid down in the EFSA Guidance (EFSA AHAW Panel, 2013); running an extensive literature search, followed by data extraction and performing a judgemental ranking exercise based on expert opinion. As main outcome, the LAPS intervention was found to be able to provide a level of animal welfare not lower than that provided by at least one of the currently allowed methods. The overall assessment of EFSA is valid ONLY under the technical conditions described in the submission and for broiler chickens, intended for human consumption, weighting less than 4 kg. Deviations from these conditions might have different consequences for animal welfare which were not assessed in this exercise. The LAPS method may, in addition to commercial slaughter, be suitable for depopulation, respecting the technical conditions defined in the present conclusions. The WG considers that a revision of the present version of the EFSA Guidance could be beneficial.

Project description:Arsenic (As) is an important environmental and food-chain toxin. We investigated the key components controlling As accumulation and tolerance in Arabidopsis thaliana. We tested the effects of different combinations of gene knockout, including arsenate reductase (HAC1), ?-glutamyl-cysteine synthetase (?-ECS), phytochelatin synthase (PCS1) and phosphate effluxer (PHO1), and the heterologous expression of the As-hyperaccumulator Pteris vittata arsenite efflux (PvACR3), on As tolerance, accumulation, translocation and speciation in A. thaliana. Heterologous expression of PvACR3 markedly increased As tolerance and root-to-shoot As translocation in A. thaliana, with PvACR3 being localized to the plasma membrane. Combining PvACR3 expression with HAC1 mutation led to As hyperaccumulation in the shoots, whereas combining HAC1 and PHO1 mutation decreased As accumulation. Mutants of ?-ECS and PCS1 were hypersensitive to As and had higher root-to-shoot As translocation. Combining ?-ECS or PCS1 with HAC1 mutation did not alter As tolerance or accumulation beyond the levels observed in the single mutants. PvACR3 and HAC1 have large effects on root-to-shoot As translocation. Arsenic hyperaccumulation can be engineered in A. thaliana by knocking out the HAC1 gene and expressing PvACR3. PvACR3 and HAC1 also affect As tolerance, but not to the extent of ?-ECS and PCS1.

Project description:Cuticular waxes are a mixture of hydrophobic very-long-chain fatty acids and their derivatives accumulated in the plant cuticle. Most studies define the role of cuticular wax largely based on reducing nonstomatal water loss. The present study investigated the role of cuticular wax in reducing both low-temperature and dehydration stress in plants using Arabidopsis thaliana mutants and transgenic genotypes altered in the formation of cuticular wax. cer3-6, a known Arabidopsis wax-deficient mutant (with distinct reduction in aldehydes, n-alkanes, secondary n-alcohols, and ketones compared to wild type (WT)), was most sensitive to water loss, while dewax, a known wax overproducer (greater alkanes and ketones compared to WT), was more resistant to dehydration compared to WT. Furthermore, cold-acclimated cer3-6 froze at warmer temperatures, while cold-acclimated dewax displayed freezing exotherms at colder temperatures compared to WT. Gas Chromatography-Mass Spectroscopy (GC-MS) analysis identified a characteristic decrease in the accumulation of certain waxes (e.g., alkanes, alcohols) in Arabidopsis cuticles under cold acclimation, which was additionally reduced in cer3-6. Conversely, the dewax mutant showed a greater ability to accumulate waxes under cold acclimation. Fourier Transform Infrared Spectroscopy (FTIR) also supported observations in cuticular wax deposition under cold acclimation. Our data indicate cuticular alkane waxes along with alcohols and fatty acids can facilitate avoidance of both ice formation and leaf water loss under dehydration stress and are promising genetic targets of interest.

Project description:Partially hydrogenated fatty acid methyl ester (H-FAME) is conventionally produced through partial hydrogenation under high pressure and elevated temperature in the presence of a catalyst. Herein, a novel green, catalyst-free, non-thermal and atmospheric pressure dielectric barrier discharge (DBD) plasma was employed instead of a conventional method to hydrogenate palm FAME. H-FAME became more saturated with the conversion of C18:2 and C18:3 of 47.4 and 100%, respectively, at 100 W input power, 1 mm gas-filled gap size and 80% H2 in the mixed gas at room temperature for 5 h, causing a reduction of the iodine value from 50.2 to 43.5. Oxidation stability increased from 12.8 to 20 h while a cloud point changed from 13.5 to 16 °C. Interestingly, DBD plasma hydrogenation resulted in no trans-fatty acid formation which provided a positive effect on the cloud point. This green DBD plasma system showed a superior performance to a conventional catalytic reaction. It is an alternative method that is safe from explosion due to the mild operating condition, as well as being highly environmentally friendly by reducing waste and energy utilization from the regeneration process required for a catalytic process. This novel green plasma hydrogenation technique could also be applied to other liquid-based processes.

Project description:RationaleThe efficiency of lubricants strongly depends on the content of functional additives. In order to assess the chemical and structural changes taking place in the lubricating oil and its additives during operation, it is essential to develop a method for simple and prompt analysis.MethodsTwo single additives as well as a fully formulated engine oil were analysed using an atmospheric pressure matrix-assisted laser desorption/ionization (AP-MALDI) source coupled to a linear trap quadrupole Orbitrap XL hybrid tandem mass spectrometer and compared with results obtained by means of electrospray ionization (ESI) including additional low-energy collision-induced dissociation (LE-CID). The identification of additives directly from technical surfaces was simulated by using steel substrates as AP-MALDI targets with varying roughness.ResultsAfter assessment and selection of the most suited AP-MALDI matrix it was found that pure additives such as calcium sulfonate and zinc dialkyldithiophosphates (ZDDPs) could well be identified with abundant signal intensity based on their elemental composition. Molecular identification was corroborated by LE-CID in ESI mode. Additionally, additives present in the fully formulated commercial oil such as ZDDPs and salicylates could be reliably identified based on the elemental composition of the deprotonated molecules by means of the Orbitrap unit on different substrates including steel surfaces with high roughness.ConclusionsAP-MALDI is an efficient technique for determination of lubricant additives directly from commercial oil blends. Identification of additive components was also achieved on steel surfaces with high roughness as applied in tribological systems and thus it is expected that it will be possible to assess additive degradation in real applications, enabling more effective and timely maintenance measures.

Project description:BackgroundA large quantity of nitrogen (N) fertilizer is used for crop production to achieve high yields at a significant economic and environmental cost. Efforts have been directed to understanding the molecular basis of plant responses to N and identifying N-responsive genes in order to manipulate their expression, thus enabling plants to use N more efficiently. No studies have yet delineated these responses at the transcriptional level when plants are grown under chronic N stress and the understanding of regulatory elements involved in N response is very limited.ResultsTo further our understanding of the response of plants to varying N levels, a growth system was developed where N was the growth-limiting factor. An Arabidopsis whole genome microarray was used to evaluate global gene expression under different N conditions. Differentially expressed genes under mild or severe chronic N stress were identified. Mild N stress triggered only a small set of genes significantly different at the transcriptional level, which are largely involved in various stress responses. Plant responses were much more pronounced under severe N stress, involving a large number of genes in many different biological processes. Differentially expressed genes were also identified in response to short- and long-term N availability increases. Putative N regulatory elements were determined along with several previously known motifs involved in the responses to N and carbon availability as well as plant stress.ConclusionDifferentially expressed genes identified provide additional insights into the coordination of the complex N responses of plants and the components of the N response mechanism. Putative N regulatory elements were identified to reveal possible new components of the regulatory network for plant N responses. A better understanding of the complex regulatory network for plant N responses will help lead to strategies to improve N use efficiency.
