Project description:Human Umbilical Vein Endothelial Cells (HUVECs) were isolated from umbilical chords by collagenase digestion and cultured to passage 3. RNA sequencing of HECTD3 knockdown during TNFalpha stimulation in primary HUVEC cells

Project description:We isolated and validated primary HUVECs from the neonatal umbilical cord vein, and performed genome-wide expression analysis to profile differentially expressed genes in control and HECTD3-KD HUVECs treated with or without TNFα for 2 hours. Compared with control HUVECs, 59 genes showed lower expression in HECTD3-KD HUVECs in response to TNFα. Most of them are NF-κB target genes, including adhesion molecules SELE (E-selectin), ICAM-1, and VCAM-1.

Effects of AEBP1 knockdown on gene expression profiles in HUVEC

Project description:We identified adipocyte enhancer binding protein 1 (AEBP1) as a novel tumor endothelial marker in colorectal cancer (CRC). To identify target genes of AEBP1 in endothelial cells, we knocked down AEBP1 in human umbilical vein endothelial cells (HUVECs). We found that genes associated with angiogenesis including aquaporin 1 (AQP1) and periostin (POSTN) are downregulated by AEBP1 knockdown in HUVECs, suggesting that AEBP1 may promote tumor angiogenesis through regulating these genes.

To delineate target genes of MEF2C in endothelial cells, which might be important during angiogenesis also, MEF2C was overexpressed adenovirally in human umbilical vein endothelial cells (HUVECs) over a period of 8 to 32 hours.
