Project description:We performed RNA sequencing of Pseudomonas syringae pv. tomato (Pst) DC3000-infected A. thaliana Col-0 and 35S::CBP60g plants at normal (23C) and elevated (28C) temperatures. 4-week-old plants were pre-incubated at 23C and 28C for 48h and then leaves were syringe-infiltrated with DC3000 bacterial inoculum. Plants were incubated at their respective temperatures (23C or 28C) for another 24h post-inoculation before tissue collection for RNA extraction. RNA samples for submitted for RNA sequencing and we found different clusters of DC3000-regulated genes that were similarly or differentially regulated between Col-0 and 35S::CBP60g at elevated temperature. Temperature-downregulated DC3000-induced genes in Col-0 plants that were restored in 35S::CBP60g plants were enriched for immunity/defense-related genes, including those essential for host salicylic acid (defense hormone) biosynthesis and accumulation.

Project description:We performed RNA sequencing of mock-inoculated and Pseudomonas syringae pv. tomato (Pst) DC3000-infected A. thaliana (Col-0 accession) plants at normal (23C) and elevated (30C) temperatures. 4-week-old Col-0 plants were pre-incubated at 23C and 30C for 48h and then leaves were syringe-infiltrated with either mock (MgCl2) or DC3000 bacterial inoculum. Plants were incubated at their respective temperatures (23C or 30C) for another 24h post-inoculation before tissue collection for RNA extraction. RNA samples for submitted for RNA sequencing and we found different clusters of DC3000-regulated genes that were downregulated, upregulated and unchanged at elevated temperature. Temperature-downregulated DC3000-induced genes were enriched for a whole suite of defense-related genes, including those essential for host salicylic acid (defense hormone) biosynthesis and accumulation.

Expression pattern in HaHB1 overexpressor plants under normal growth conditions

Project description:Plants deal with cold temperatures via different signal transduction pathways. To elucidate the molecular mechanisms by which HaHB1 conferred tolerance to cold temperatures, we performed an exploratory transcriptome analysis comparing RNAs of 3-week-old transgenic and WT plants. Several transcripts encoding glucanases and chitinases were induced. Notably, there was not a considerable overlap with CBF regulon genes previously described in cold tolerance molecular mechanism. We propose that HaHB1 is involved in plant cold tolerance via the induction of proteins able to stabilize cell membranes and inhibit ice growth.

Global gene expression of Arabidopsis lines with altered ANAC102 expression under normal and low oxygen conditions

Project description:Global gene expression was compared between Arabidopsis lines with altered expression of ANAC102 (over-expressed and knocked-out) and wild-type. ANAC102 is a putative NAC domain transcription factor. Gene expression was compared between an ANAC102 over-expressing line and parental ecotype C24 under ambient atmosphere to determine which genes ANAC102 is capable of regulating. Gene expression was also compared between three week old plants of an ANAC102 knock-out line and parental ecotype Col-0 under 0.1% Oxygen and ambient atmosphere conditions to determine which genes may require ANAC102 for appropriate expression under these conditions. Global gene expression analysis of Arabidopsis Col-0 under normal and low oxygen conditions

Project description:Global gene expression was compared between root RNA samples from three-week-old Arabidopsis Col-0 plants subjected to 0.1% oxygen (balance nitrogen) or ambient atmospheric conditions. Three-week-old Arabidopsis seedlings (4-6 leaves) of ecotype Col-0 were used. Two biological replicates were used for each treatment, and for each biological replicate, five Arabidopsis seedlings grown on the same petri dish were bulked. RNA was extracted from root tissue excised post-treatment. Treatment consisted of moving plants grown on solid MS media containing 3% sucrose to liquid MS media containing 3% sucrose for 24 hours, after which plates were placed in anaerobic cannisters covered in aluminum foil. Cannisters containing plants in the 0.1% oxygen treatment group were flushed with a 0.1% oxygen gas mixture and then sealed, while cannisters in the control group were left unsealed. Treatment lasted 5 hours before plant tissue was harvested and RNA extracted.

Transgenic plants used for experiment were: overexpressor plants obtained by transformation of Col-0 WT plants with genetic constructs created in [Tarasenko et al., 2016] contained catalytic part of RPOTmp enzyme with transit peptides of RPOTm (mitochondrial) and RPOTp (plastid) by agrobacterial transformation; plants with complementation of RPOTmp functions in mitochondria or chloroplasts obtained from transformation of GABI_286E07 rpotmp knockout-mutant plants with genetic constructs created in [Tarasenko et al., 2016]. Goal was to determine the effects of RPOTmp knockout/overexpression on global Arabidopsis thaliana gene expression.
