Expression data of tumor tissues and tumor sphere from neuroblastoma patients

ABSTRACT: Cancer stem cells (CSCs) are regarded as the starting point for tumors and are considered to play key roles in relapse and metastasis in many cancers include neuroblastoma (NB). CSCs have become promising target cancer cells for preventing cancer relapse and improving the prognosis of tumor patients. The CSCs marker are not established in NB, but CSCs are able to isolated as tumor sphere. We used a microarray to profile the transcriptomes of NB tumor spheres to select CSCs candidate molecules.

Altered expression of microRNAs in PC-3 sphere cells of prostate cancer compared with PC-3 adherent cells

Project description:Metastatic prostate cancer is a leading cause of cancer-related death in men. Cancer stem cells (CSCs) are involved in tumor progression and metastasis, including in prostate cancer. There is an obvious and urgent need for effective cancer stem cells specific therapies in metastatic prostate cancer. MicroRNAs (miRNAs) are an important class of pervasive genes that are involved in a variety of biological functions, especially in cancer. The goal of this study was to identify miRNAs involved in prostate cancer metastasis and cancer stem cells. Several published reports have demonstrated that non-adherent spheres culture is increasingly used as an effective method to enrich and identify stem cells or putative CSCs.In our previous study, we enriched prostate cancer stem cells from PC-3 sphere cells in serum-free suspension culture and characterized their CSCs properties.Thus, we used spheres as a prostate cancer stem cells model to elucidate its metastatic mechanisms. We examined the miRNA expression profiles of PC-3 sphere cells of prostate cancer compared with PC-3 adherent cells by miRNA microarray.

Identify the cancer stem cells expression characteristics in ovarian and breast cancer

Project description:The identification and characterization of subpopulations of cancer stem cells (CSCs) provide new understandings and possible therapeutic implications in cancer biology. We found the ovarian cancer sphere cells possessed CSCs properties maintained self renewal, drug resistance, and tumorigenesis. Using high-throughput microarray system, we identified common GO terms and pathway signatures significantly enriched in ovarian and breast cancer stem cells. Ovarian and breast cancer cells were cultured in sphere formation conditions, and total RNA from those spheres and conresponding adhered cell was hybridized on Affymetrix microarrays.

Gene expression of clinical breast cancer cells cultured by sphere and adherent culture methods

Project description:Cancer stem-like cells (CSCs) are thought to be responsible for recurrence of tumors and poor prognosis because of their higher tumor initiating ability and drug resistance, though the mechanisms remain still unclear. Here, we show a critical role of mini-chromosome maintenance protein (MCM) 10, a component of DNA replication machinery, in enabling CSCs to deal with DNA replicative stress. Our transcriptomic analysis of patient-derived cultured breast cancer cells revealed that MCM10 is strongly upregulated in CSC-enriched spheroid cells, compared to cancer cells cultured in a regular condition. Elevated MCM10 expression was correlated with worse prognosis of patients with breast cancer. Depletion of MCM10 resulted in not only reduced cell proliferation and tumorigenesis, but also reduced tumor sphere forming efficiency, expression levels of stemness markers, and the tumor initiating ability. These results indicate that MCM10 is essential for maintenance of CSCs as well as for tumor cell proliferation. DNA fiber assay revealed that CSCs have a comparatively high level of DNA replicative stress. Overexpression of MCM10 helped cells to proliferate in a medium containing hydroxyurea (HU), a replicative stress-inducing chemotherapeutic agent. Conversely, MCM10 depletion made cells more fragile to replicative stress induced by HU or mitomycin C (MMC), another replicative stress inducer. Therefore, MCM10 plays critical roles for dealing with the enhanced replicative stress generated in CSCs for their survival, leading to maintenance of tumor initiating ability and drug resistance. We provide a preclinical rationale to target MCM10, the critical mediator for DNA replicative stress, for breast cancer that includes CSCs.

Development of gene expression signatures for ovarian cancer stem cells

Project description:To understand the differentiation of ovarian cancer stem cells (CSCs), We derived two phenotypes of CSCs and identified the gene expression profiling. The CSCs were derived from Cp70 ovarian cancer cells and cultured in suspension and examined every day for sphere formation. Spheres were then dissociated and passaged at least eight times in 2 months to generate spheres, which are henceforth referred to as SR cells. The surface of SR-I was smooth regardless of the size, whereas SR-II was morula. SR-I could differentiate into multiple-lineage cell types under specific induction conditions. SR-I spheroids could differentiate to SR-II spheroids through epithelial-mesenchymal transition.The self-renewal was slower for SR-I than for SR-II.

Transcriptome analysis of control and SKOV3 floating sphere cells and sphere differentiated cells

Project description:We initiated this study with the aim of systematically identifying the biomarkers driving tumorigenesis and maintaining tumor dedifferentiation in human ovarian cancer. To achieve that goal, SKOV3 cells were seeded in ultra-low-attachment 96-well plates in DMEM/F12 medium supplemented with 2% B27 serum replacement to allow the formation of floating spheres. Then, these spheres were collected and recultured in a 6-well plate in DMEM with 10% FBS to allow the cells in the spheres to differentiate and attach to the bottom surface, and the genes in which transcription patterns were altered during the transition from OCSCs to differentiated cells.

CD73 as a therapeutic target of pancreatic neuroendocrine tumor stem cells

Project description:Background: Identification and purification of cancer stem cells (CSCs) lead to new therapeutic targets; however, there has been no study to identify and isolated pancreatic neuroendocrine tumor (pNET) CSC. Therefore the clinical significance and its target remain unknown. This study aimed to identify pNET CSCs and characterize therapeutic candidate for pNET CSCs. Methods: We isolated CSCs sorting by ALDH activity in pNET surgical section and cell lines. We verified whether these cells have the property of stemness in vivo and in vitro. Additionally in order to acquire CSC gene profile, genome-wide gene expression profiles were investigated using a microarray technique. Results: ALDHhigh cells, but not control bulk cells, formed spheres, proliferated in hypoxia as well as normoxia and promoted cell motility, which are features of CSCs. Injection of as few as 10 ALDHhigh cells led to subcutaneous tumor formation, and 105 ALDHhigh cells established metastases but not control bulk cells in mice. Comprehensive gene expression analysis revealed that genes associated with mesenchymal stem cell, including CD73, and epithelial-mesenchymal transition (EMT) were overexpressed in ALDHhigh cells. APCP, which is CD73 inhibitor, inhibited sphere formation and cell motility in ALDHhigh cells in vitro, and tumor growth inhibition were observed in ALDHhigh cells in vivo. Conclusions: We identified ALDHhigh cells of pNET and elucidated that they have stemness property. Furthermore we identified CD73 as a target of ALDHhigh cells. CD73 is a promising novel target of pNET CSCs.

Tumorspheres from transformed fibroblasts highlight the great plasticity of differentiated tumorigenic cells

Project description:Tumors are heterogeneous tissues. A subpopulation of cells with stemness features (cancer stem cells: CSCs) has been implicated in tumor maintenance and progression. There is evidence that CSCs can originate from tumor cell dedifferentiation. Here we used in vitro transformed human fibroblasts (cen3tel cells) and the tumorsphere assay to search for and possibly characterize CSCs from differentiated transformed cells. To obtain tumorspheres, cen3tel cells were grown in the absence of serum and in the presence of growth factors. Sphere cells were analysed for the expression of specific genes and their genome wide gene expression profiles relative to cells grown in adhesion were determined by microarray analysis. Cen3tel cells formed spheres showing self-renewal capacity and Sox2 overexpression, suggesting that they contained a subset of cells with CSC-like features. Sphere cells displayed the deregulation of a c-MYC-miR-34a circuitry, likely associated with cell protection from apoptosis. Gene expression profiles of sphere cells revealed an extensive transcriptional reprogramming. Genes upregulated in tumorspheres identified processes related to tumorigenesis and stemness, such as chromatin organization, cholesterol biosynthetic pathway, apoptosis suppression, immunity and defense, interferon mediated immunity and cytokine and chemokine mediated signalling pathway. However, sphere cells were only slightly more tumorigenic in vivo than adherent cells. Our results indicate that tumorigenic cells obtained from differentiated human fibroblasts are highly plastic and can acquire cancer stem cells properties. However, tumorsphere cells are not the main tumorigenic population, highlighting a further level of complexity in tumor heterogeneity.
