Project description:The transport of auxin controls the rate, direction and localization of plant growth and development. The course of auxin transport is defined by the polar subcellular localization of the PIN proteins, a family of auxin efflux transporters. Using blue native PAGE and quantitative mass spectrometry, we identify native PIN core transport units as homo- and heteromers assembled from PIN1, 2, 3, 4 and 7 subunits only. To identify the protein composition of the larger, low stringency-solubilized PIN assembly we affinity-purified GFP-fused PIN proteins from Arabidopsis roots after expression from their native promoters and after the induction of discrete lateral roots, and analyzed the captured proteins by label-free quantitative mass spectrometry (LC-MS/MS).

Project description:To evaluate the role of seeds in fruit quality, we induced parthenocarpy in tomato by regulating ovule-specific auxin synthesis or responsiveness using the INO promoter from A. thaliana, which is expressed in the outer layer of the integuments during early stages of ovule development. We compared these to fruit where the same coding regions were expressed from the DeFH9 promoter which is expressed in carpel tissues during early stages of ovule development. Expression of auxin synthesis or responsiveness genes by both of these promoters produced seedless parthenocarpic tomato fruit. We compared fruit samples using the Affymetrix tomato GeneChip (GPL4741) to determine how gene regulation and expression differed between wild-type and transgenic fruit. Project description:Auxin is a pivotal plant hormone that controls virtually all aspects of plant growth and development. Auxin is transported in plants in a polar fashion and PIN-FORMED (PIN) transporters are the key players in this process. Canonical PINs possess a long cytosolic loop that is subject to regulation by AGC1 and AGC3 kinases. To characterize the transporters, they are expressed in Xenopus laevis oocytes alone or with the different activating kinases. Since transport rates always depend on the amount of active transporter in the membrane it is paramount to ensure that the levels of transporter, kinase and degree of phosphorylation are similar to be able to compare transport rates directly. To this end we expressed PIN1, PIN2 and PIN3 either alone or with D6PK (an AGC1 kinase) or PID (an AGC3 kinase) in oocytes. The protein levels and the degree of phosphorylation of the loop were established and found to be comparable between PINs.

Project description:D6 PROTEIN KINASE (D6PK) is a polarly localized plasma membrane-associated kinase from Arabidopsis thaliana that activates polarly distributed PIN-FORMED (PIN) auxin transporters. D6PK traffics rapidly to and from the plasma membrane, independently from its PIN targets. D6PK plasma membrane association is dependent on the middle domain, an insertion between kinase subdomains VII and VIII. How D6PK polar plasma membrane targeting is established and maintained remains to be understood. Here, we show that cysteines of repeated CXX(X)P motifs in the middle domain are S-acylated and required for D6PK membrane association. While D6PK S-acylation is not detectably regulated during intracellular trafficking, the phosphorylation of adjacent serine residues, in dependence of the upstream regulatory 3-PHOSPHOINOSITIDE-DEPENDENT PROTEIN KINASES1, regulates D6PK residence time at the plasma membrane, lateral diffusion, and trafficking. We describe a mechanism for the regulation of D6PK plasma membrane interaction and polarity, which may also explain the polarity regulation of related kinases.

Experiment Overall Design: Wild-type fruit with seeds was compared with transgenic lines INO-IaaM, DefH9-IaaM, INO-RolB, and DefH9-RolB. To find genes with seed-specific expression, we also compared the control with wild-type fruit from which seeds had been manually removed. We had three biological replicates for each treatment and control except DefH9-RolB, for which only two replicates were available. Each CEL file from the microarray represents one plant from each line.

Project description:Li-Fraumeni syndrome (LFS) is a hereditary cancer predisposition syndrome associated with a highly penetrant and diverse tumor spectrum characterized by germline mutations in the TP53 tumor suppressor gene. To better understand how TP53 mutations predispose individuals with LFS to cancer development, we characterized tp53 R217H and R242H zebrafish lines as the first zebrafish p53 hotspot mutants (human R248 and R273H). These mutants have lost several key wildtype p53 functions and recapitulate many LFS phenotypes. Specifically, we have shown that the R217H and R242H alleles result in partial-to-no activation of key p53 target genes, are resistant to apoptosis in a dominant negative manner and exhibit a defective G1 cell-cycle checkpoint in vivo. The loss of these wildtype p53 functions predisposed the fish to develop spontaneous tumors as early as 6 months of age. Tumor histology resembles human sarcomas, a predominant LFS tumor type. tp53 R242H mutants developed tumors earlier with a higher lifetime incidence than tp53 null or R217H mutants, suggesting it is a more aggressive mutation, while the R217H allele may be hypomorphic. Additionally, we observed mutation-specific differences both in the tumor type and sex bias across tp53 null, R217H, and R242H genotypes with associated diverse transcriptomic and DNA methylome profiles, impacting metabolism, cell signalling, and biological macromolecule synthesis and degradation. These tp53 zebrafish mutants demonstrate fidelity to their human counterparts and provide new insights into underlying tumorigenesis mechanisms and kinetics, which may inform more tailored tumor surveillance approaches and novel therapeutic targets in LFS.
