Project description:The cyanobacterial marine natural product honaucin A inhibits mammalian innate inflammation in vitro and in vivo. To decipher its mechanism of action, RNA sequencing was used to evaluate differences in gene expression of cultured macrophages following honaucin A treatment. This analysis led to the hypothesis that honaucin A exerts its anti-inflammatory activity through activation of the cytoprotective nuclear erythroid 2-related factor 2 (Nrf2)-antioxidant response element/electrophile response element (ARE/EpRE) signaling pathway. Activation of this pathway by honaucin A in cultured human MCF7 cells was confirmed using an Nrf2 luciferase reporter assay. In vitro alkylation experiments with the natural product and N-acetyl-l-cysteine suggest that honaucin A activates this pathway through covalent interaction with the sulfhydryl residues of the cytosolic repressor protein Keap1. Honaucin A presents a potential therapeutic lead for diseases with an inflammatory component modulated by Nrf2-ARE.

Project description:Secondary injury causes death and dependence after spontaneous intracerebral haemorrhage (ICH). We found that myelomononuclear Nrf2 is active and protective after ICH in mice. To investigate the roles of myelomononuclear Nrf2 in modulating cell autonomous and non-cell autonomous responses to the oxidative and inflammatory consequences of ICH we developed an in vitro system of co-cultures of primary mouse microglia, human astrocytes and rat neurons and performed RNA-seq, subsequently performing in-silico separation of read data into separate species, and thus separate cell types. Cultures were treated with blood clot condition media (CCM), lipopolysaccharide (LPS), and Nrf2-activating drug CDDO-TFEA, and experiments were performed using wild-type mouse microglia, and microglia from mice with global Nrf2 deletion.

Project description:LncRNA for anti-oxidative capacity control has not been discovered yet. Nrf2 is a central molecule for cellular defense by increasing anti-oxidative capacity. Here, we identified a novel lncRNA named ‘NACER’ (Nrf2 Activating Competing Endogenous RNA) transcribed from an upstream region of MIR122. NACER existed in the cytoplasm, suggestive of its function as competing endogenous RNA (ceRNA, miRNA sponge). NACER served as ceRNA for Plk2 and p21cip1 through binding for miR-128/224, inducing Plk2/Nrf2/p21cip1 complexation for Nrf2 activation in cells under p53-activating conditions (i.e., DNA damage and serum deprivation). NACER expression was suppressed when apoptosis was initiated, being a rheostat for cell fate determination. NACER levels correlated with the recurrence free survival rate of patients with hepatocellular carcinoma after surgery. Collectively, NACER promotes Plk2 and p21cip1 translation by competing for specific miRNAs, activating Nrf2 under the surviving condition from oxidative stress, implying that NACER plays a fine-tuning rheostat for cell fate decision.

Project description:Nrf2 (NF-E2-related-factor-2) contributes to the maintenance of glucose homeostasis in vivo. Nrf2 suppresses blood glucose levels by protecting pancreatic β-cells from oxidative stress and improving peripheral tissue glucose utilization. To elucidate the molecular mechanisms by which Nrf2 contributes to the maintenance of glucose homeostasis, we generated skeletal muscle (SkM)-specific Keap1-knockout (Keap1MuKO) mice that express abundant Nrf2 in SkM and then examined Nrf2-target gene expression in this tissue. In Keap1MuKO mice, blood glucose levels were significantly downregulated, and the levels of glycogen branching enzyme (Gbe1) mRNA, along with those of glycogen branching enzyme (GBE) protein, were significantly upregulated in mouse SkM. Consistent with this result, chemical Nrf2-inducers promoted Gbe1 mRNA expression in both mouse SkM and C2C12 myotubes. Chromatin-immunoprecipitation analysis demonstrated that Nrf2 binds the Gbe1 upstream promoter regions. In Keap1MuKO mice, muscle glycogen content was strongly reduced, and forced GBE expression in C2C12 myotubes promoted glucose uptake. Therefore, our results demonstrate that Nrf2-induction in SkM increases GBE expression and reduces muscle glycogen content, resulting in improved glucose tolerance. Project description:Marine natural products (MNPs) are recognized for their structural complexity, diversity, and novelty. The vast majority of MNPs are pharmacologically relevant through their ability to modulate macromolecular targets underlying human diseases. Angiogenesis is a fundamental process in cancer progression and metastasis. Targeting angiogenesis through selective modulation of linked protein kinases is a valid strategy to discover novel effective tumor growth and metastasis inhibitors. An in-house marine natural products mini-library, which comprises diverse MNP entities, was submitted to the Lilly's Open Innovation Drug Discovery platform. Accepted structures were subjected to in vitro screening to discover mechanistically novel angiogenesis inhibitors. Active hits were subjected to additional angiogenesis-targeted kinase profiling. Some natural and semisynthetic MNPs, including multiple members of the macrolide latrunculins, the macrocyclic oxaquinolizidine alkaloid araguspongine C, and the sesquiterpene quinone puupehenone, showed promising results in primary and secondary angiogenesis screening modules. These hits inhibited vascular endothelial growth factor (VEGF)-mediated endothelial tube-like formation, with minimal cytotoxicity at relevant doses. Secondary kinase profiling identified six target protein kinases, all involved in angiogenesis signaling pathways. Molecular modeling and docking experiments aided the understanding of molecular binding interactions, identification of pharmacophoric epitopes, and deriving structure-activity relationships of active hits. Marine natural products are prolific resources for the discovery of chemically and mechanistically unique selective antiangiogenic scaffolds.
