Project description:Purpose : This study was performed to investigate the effects of knockdown of PPARg for osteo/cementogenic differentiation of PDLF through RNA sequencing. Method : PDLF were transfected wtih either control siRNA or siRNA for PPARg and then cultured in mineralization inducing medium for 6 days. Total RNAs were collected before and after the culture. Result : Gene ontology analyses of RNA-seq data revealed the genes functionally related with ECM such as collagen synthesis and trimerization, proteoglycans, and fibers were selectively increased by 6-day culture and suppressed biological processes by si-PPARg compared with si-control at day 6 exhibited 5 of 7 upper ranked biological processes were related with ECM organization and metabolism. Moreover, the upper rank of suppressed pathways by si-PPARg compared with si-control at day 6 showed `Endochondral Ossification` was ranked 1st. Conclusions:These results suggested that knockdown of PPARg was sufficient to eliminate osteo/cementogenic and ECM organizing potentials by comprehensive transcriptional dysregulation in PDLF.

Project description:Periodontal ligament is soft connective tissue between alveolar bone and cementum, the surface hard tissue of tooth. Periodontal ligament fibroblasts (PDLF) actively express osteo/cementogenic genes, which contribute in periodontal tissue homeostasis. However, the key factors by which PDLF retain such osteo/cementogenic abilities has not been clarified yet. Here, we find PPARg is expressed in vivo periodontal ligament tissue and its distribution pattern is highly correlated with the alkaline phosphate enzymatic activity. Knockdown of PPARg drastically deprives PDLF of osteo/cementogenic abilities in vitro. In contrast, PPARg agonists enhance it. PPARg is required for maintaining acetylation status of H3K9 and H3K27, active chromatin marks, and the supplementation of acetyl-coA, the donor of histone acetylation, is able to restore PPARg knockdown-induced decreased osteo/cementogenic abilities in PDLF. RNA-seq and ChIP-seq combined analyses identify 4 osteogenic transcripts, RUNX2, SULF2, RCAN2, and RGMA, in the PPARg-dependent active chromatin region marked by H3K27ac. Furthermore, the RUNX2 binding sites are selectively enriched in the PPARg-dependent active chromatin region. Together, these findings identify PPARg is the key transcriptional factor of PDLF to retain osteo/cementogenic abilities and reveal that global H3K27ac modification involves in the comprehensive osteo/cementogenic transcriptional alteration mediated by PPARg.

Project description:In this study, the effect of a storage medium (hK-HTCM) in which hair keratin was dissolved in a 1:1 mixed solution of Histidine-Tryptophan-Ketoglutarate and Culture media solution (HTCM) was evaluated on the viability and proliferation of human periodontal ligament cells. There was no difference in cytotoxicity between 0.1% and 0.25% hK-HTCM against 0% hK-HTCM and human periodontal ligament cells. Human periodontal ligament cells were cultured in 0.1% and 0.25% hK-HTCM for 48 hours, and after removing hair keratin from the culture medium, the cells resumed proliferation. When exposed to 0.25% hK-HTCM, human periodontal ligament cells showed differential expression of genes related to cell cycle and cell division regulation. On the other hand, differential expression of genes related to phosphorylation and ubiquitination related to cell cycle resumption was observed in human periodontal ligament cells after removal of 0.25% hK-HTCM. 0.25% hK-HTCM showed the ability to regulate the cell cycle of human periodontal ligament cells without showing cytotoxicity, and its potential to be used as a long-term storage medium for avulsed teeth was confirmed.

Project description:The structural and functional integrity of bone-periodontal ligament (PDL)-cementum complex stems from the load-bearing attachment sites (entheses) between soft (PDL) and hard (bone, cementum) tissues. These attachment sites are responsible for the maintenance of a bone-PDL-cementum complex biomechanical function. The objective was to investigate changes in spatiotemporal expression of key biomolecules in developing and functionally active entheses.Multilabeling technique was performed on hemimandibles of 3 wk and 3 mo-old scleraxis-GFP transgenic mice for CD146, CD31, NG2, osterix and bone sialoprotein. Regions of dominant stretch within the PDL were evaluated by identifying directionality of collagen fibrils, PDL fibroblasts and PDL cell cytoskeleton.CD146+ cells adjacent to CD31+ vasculature were identified at PDL-bone enthesis. NG2+ cells were located at coronal bone-PDL and apical cementum-PDL entheses in the 3-wk-old group, but at 3 mo, NG2 was positive at the entheses of the apical region and alveolar crest. NG2 and osterix were colocalized at the osteoid and cementoid regions of the PDL-bone and PDL-cementum entheses. Bone sialoprotein was prominent at the apical region of 3-wk-old mice. The directionality of collagen fibers, fibroblasts and their cytoskeleton overlapped, except in the apical region of 3 wk.Colocalization of biomolecules at zones of the PDL adjacent to attachment sites may be essential for the formation of precementum and osteoid interfaces at a load-bearing bone-PDL-tooth fibrous joint. Biophysical cues resulting from development and function can regulate recruitment and differentiation of stem cells potentially from a vascular origin toward osteo- and cemento-blastic lineages at the PDL-bone and PDL-cementum entheses. Investigating the coupled effect of biophysical and biochemical stimuli leading to cell differentiation at the functional attachment sites is critical for developing regeneration strategies to enable functional reconstruction of the periodontal complex.

Project description:Loss of tissue attachment as a consequence of bacterial infection and inflammation represents the main therapeutic target for the treatment of periodontitis. Cementoblasts, the cells that produce the mineralized tissue, cementum, that is responsible for connecting the soft periodontal tissue to the tooth, are a key cell type for maintaining/restoring tissue attachment following disease. Here, we identify two distinct stem cell populations that contribute to cementoblast differentiation at different times by lineage tracing and single cell RNA sequencing. During postnatal development, cementoblasts are formed from perivascular‐derived cells expressing CD90 and perivascular‐associated cells that express Axin2. During adult homeostasis, only Wnt‐responsive Axin2+ cells form cementoblasts but following experimental induction of periodontal disease, CD90+ cells become the main source of cementoblasts. We thus show that different populations of resident stem cells are mobilized at different times and during disease to generate precursors for cementoblast differentiation and thus provide an insight into the targeting cells resident cells for novel therapeutic approaches.

Further biomolecular study has suggested that the expression of FDC-SP may be associated with periodontal ligament (PDL) phenotype expression. Therefore, we hypothesized that FDC-SP may play specific roles in the inhibition of calcium precipitation during periodontal regeneration, as well as affect phenotype expression of periodontal ligament cells (PDLCs) during the differentiation process. To investigate this, we applied microarray technology to identify gene expression changes in hPDLCs transfected with FDC-SP and then clustered them according to their biological functions.

Project description:Cancer is a rapidly evolving, multifactorial disease that accumulates numerous genetic alterations. This results in phenotypic and molecular heterogeneity within the tumor, the complexity of which is further amplified through specific interactions between cancer cells. We aimed at dissecting the molecular mechanisms underlying the cooperation between different clones. We produced clonal cell lines derived from the MDA-MB-231 breast cancer cell line, using the UbC-StarTrack system, which allows tracking of multiple clones by colour: GFP C3, mKO E10 and Sapphire D7. Characterization of these clones in vitro revealed clear differences at genetic levels, that induce differences in growth rate, morphology and cytokine expression among them. In vivo, all clonal cell lines were able to form tumors, however an injection of an equal mix of clones, led to the formation of tumors where mKO E10 was almost depleted. Additionally, the mKO E10 clonal cell line showed a significant deficiency in the formation of lung metastasis. These results confirm that even in stable cell lines heterogeneity is present. In vitro the complementation of growth medium with medium or exosomes from either parental or clonal cell lines, increased the growth rate of the other clones. Co-growth and co-injection of mKO E10 and GFP C3 clonal cell lines increased the efficiency of invasion and migration, respectively. These findings support a model where the interplay of clones confers aggressiveness and may allow to identify factors involved in cellular communication which can be involved in clonal cooperation and be new targets preventing tumor progression.

After that, we applied Agilent Whole Human Genome Oligo Microarray (4M-CM-^W44K) to identify differentially expressed genes between empty vector-transfected hPDLCs and FDC-SP -transfected ones and then clustered them according to their biological functions. 3 independent experiments were performed and the empty vector-transfected hPDLCs were used as control.
