ABSTRACT: The dataset contains data from 9 samples hybridized on Illumina HumanHT-12 array. -patient's primary lymphoma cells obtained from leukemized blood (P7-PBMC) -non-malignant B-cells isolated from peripheral blood of 5 healthy donors (CTRL) -lymphoma cell line named UPF1G established from the patient's primary lymphoma cells before cytarabine-based therapy after 63 and 163 days of in vitro cultivation (UPF1G-D63, UPF1G-D163) -lymphoma cell line named UPF1H established from the pateint's primary lymphoma cells after cytarabine-based therapy after 85 and 185 days of in vitro cultivation (UPF1H-D85, UPF1H-D185) -cells established by xenotransplantation of UPF1G cell line isolated ex vivo from subcutaneously growing lymphoma (UPF1G-SC) -cells established by xenotransplantation of UPF1G cell line isolated ex vivo from malignant murine ascites (UPF1G-A) -cells established by xenotransplantation of UPF1H cell line isolated ex vivo from subcutaneously growing lymphoma (UPF1H-SC) Samples were sorted using CD19-microbeads (Miltenyi). For xenotransplantation, NOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJ mice (referred to as NSG mice) were used.

A bladder cancer patient-derived xenograft reflects aggressive growth dynamics in vivo and in organoids

Project description:A MIBC patient with aggressive disease course was selected for ex vivo model establishment and growth dynamic studies. After standard neoadjuvant treatment with gemcitabine and cisplatin, cystectomy was performed, and excised urothelial carcinoma tissue was implanted subcutaneously into male SCID mice to generate CoCaB1 PDXs. Established PDXs were passaged subcutaneously in SCID mice to generate early and late passage models for functional and molecular characterization. Representative early and late passage PDXs were collected for organoid establishment.

Proteomics of Mouse Syngeneic Models

Project description:The cell line-derived xenografts and patient derived xenografts have limited use in cancer immunotherapy evaluation because an immune compromised host is required for xenotransplantation. Syngeneic mouse models are derived by transplanting established mouse cell lines or tumor tissues to strain matched mouse hosts, which are better suited to study the interplay between immune and tumor cells. We investigated the differences as well as similarities of a panel of ten mouse syngeneic models to features of human tumors by proteomics, which will provide valuable information to assist experimental biologists in model selection.

Effect of FL-Fc-DM1 drug on gene expression of THP-1 cells pretreated with cytarabine.

Project description:Cytarabine is one of the first line chemo drugs to treat acute myeloid leukemia (AML). However, cytarabine can induce AML cell cell cycle arrest at G0/G1 phase. These cell-cycle-arrested AML cells resist cytarabine, show a senescence-like phenotype or stem-cell like phenotype. To eliminate these cell-cycle arrested cells, we developed a FL-Fc-DM1 drug by conjugating FLT3 liangd Fc fusion protein and mertansine. We used the THP-1 cells as a AML model. The total mRNA of THP-1 cells treated with different drugs (cytarabine, FL-Fc-DM1, FL-Fc) were sequenced.
