Ectoderm and mesenchyme gene expression in the developing mouse face

Project description:This investigation provides a robust multi-dimensional compendium of gene expression data relevant to mouse facial development. It profiles the transcriptome ofectoderm and mesenchyme from the three facial prominences in a time series encompassing their growth and fusion. Analysis of the dataset identified more than 8000 differentially expressed genes comprising dramatically different ectoderm and mesenchyme programs. The mesenchyme programs included many genes identified in earlier analyses as well hundreds of genes not previously implicated in craniofacial development. The ectoderm programs included over a thousand genes that highlight epithelial structure, cell-cell interactions and signaling. The dataset includes 45 .cel files, DABG probability and RMA log2 expression values for each probeset, and statistics for 9457 probesets representing 8575 genes. 45 total samples, with 15 conditions sampling three ages (E10.5, E11.5, E12.5), three facial prominences (mandibular, maxillary and fronto-nasal) and two tissue layers (ectoderm or mesenchyme), with 3 biological replicates per condition. Identification of transcriptome of mouse non-neural ectoderm during nueral tube closure and gene enrichment compared to remaining neural tube tissue

Project description:The process of neural tube closure is a highly complex morphogenetic event that results in the generation of the primordial central nervous system. During formation of the neural tube, the non-neural ectoderm separates from the neighboring neural ectoderm and forms a single layer epithelial sheet that overlies the closed neural tube. Previous work has shown that the non-neural ectoderm is necessary for proper cranial neural tube closure, however little is known about this cell population at the molecular level or how the non-neural ectoderm contributes to neural tube closure. In this study, we used a mouse genetic system to fluorescently label the non-neural ectoderm cells and FACS sorted these cells away from the other cell populations in the neural tube. We performed high throughput RNA-sequencing to identify the transcriptome of the non-neural ectoderm and compared the gene expression profile of non-neural ectoderm cells to the remaining population of cells within the neural tube in order to identify which genes are enriched within the non-neural ectoderm. This analysis provides a clue as to which underlying molecular processes may be important for non-neural ectoderm function during neural tube closure.

Transcriptional profiling to compare gene expression in supraorbital mesenchyme (SOM) and early migration mesenchyme (EMM) of wild type mouse embryos

Project description:During embryonic development, the mesenchyme surrounding the brain (cranial mesenchyme) gives rise to several structures such as the meninges, calvaria, and the dermis of the scalp. It has been known that the two regions within the cranial mesenchyme, namely, SOM on the basolateral side and EMM on the apical side of the head, have different developmental fates. However, the molecular genetic basis underlying this difference had been unclear. To address this question, we sought to identify genes differentially expressed between EMM and SOM. Through RNAseq, we identified 1095 genes with higher expression in EMM than in SOM, and 398 genes with higher expression in SOM than in EMM (fold change>2, p<0.001, base mean count>50).
