Gene expression in Peyronie disease, tunica albuginea, and corpora cavernosum in cultured cells from human penis

ABSTRACT: Cells from normal tissues and from a Peyronie Disease plaque of human penis were cultured and RNA was extracted. The aim was to investigate differences in gene expression in the abnormal plaque as well as to establish baseline expression data in normal tissues for comparison.

An extra-genital cell population contributes to urethra closure during mouse penis development

Project description:Penis, the organ that bears reproductive and psychological importance, is susceptible to birth defects such as hypospadias, or incomplete closure of urethra along the penis shaft. We discover that proper urethral closure in mouse embryos requires a unique mesenchymal cell population originated from outside of the penis. These “extra-genital” cells, marked by a lineage marker Nr5a1, migrate from the inguinal region into the embryonic penis, and facilitate urethra closure by interacting with adjacent peri-urethral cells via the epidermal growth factor pathway. Ablation of Nr5a1+ cells leads to severe hypospadias, and alters cell differentiation in the penis. This discovery highlights the indispensable role of Nr5a1+ extra-genital cells in urethra closure, shedding light on the biology of penis formation and potential implications for human hypospadias.

Landscape of microenvironment in Randall's plaque by single-cell sequencing

Project description:Randall's plaque is significantly associated with occurrence of nephrolithiasis. However, the microenvironment of Randall's plaque is poorly characterized. To investigate the microenvironment of Randall's plaque, we analyzed single-cell RNA data of 3 Randall's plaque and 3 normal renal papillae tissue and identified 11 different cell types. We screened differentially expressed genes among all cell types between Randall's plaque and normal renal papillae. The microenvironment showed two cell types with multiple stone formation-associated transcriptomic programs. Contrary to previous studies, we did not observe macrophages M1/M2 imbalance. Notably, we detected ossification-associated macrophage is enrich in Randall's plaque and validated GPNMB and ACP5 were potential biomarkers on osteoblasts-associated macrophage. We also identified an endothelial subset harboring active communication (COL15A1+ PCDH17+ endothelial, DPECs) with other cells. Together with Immunofluorescence, we validated ossification-associated macrophage and DPECs are enriched in Randall's plaque tissue. Finally, cell-to-cell communication revealed that Loop of Henle, DPECs and osteoblasts-associated macrophages were the main source of SPP1 signaling. Our work will further the understanding of the microenvironment among Randall's plaque tissues and provide deep insight into immune modulation.

Project description:Peyronie’s disease (PD) is a severe fibrotic diseases in the tunica albuginea that causes penis curvatures and leads to penile pain, deformity as well as erectile dysfunction (ED). The role of pericytes in the pathogenesis of fibrosis have been perceived recently. Extracellular vesicles (EVs)-mimetic nanovesicles (NVs) have emerged particular interest in intercellular communication between cells in various fields of fibrosis disease. However, the global gene expression of pericytes-derived EVs-mimetic NVs in regulating fibrosis remains unknown. Here we used RNA-sequencing technology to investigate the potential target genes regulated by pericytes-derived EVs-mimetic NVs in primary fibroblasts derived from human PD plaque.

Inflammatory pseudotumor of the tunica albuginea and the tunica vaginalis: Case-report.

Project description:Inflammatory pseudo tumor of the tunica is rare and typically presents as long standing, painless scrotal mass. A 23-year-old man had palpable, multiple, hard scrotal masses for 3 months. Laboratory investigations were normal (including LDH, AFP, HCG). Radical inguinal orchiectomy done. Macroscopically the testis and epididymis were normal, with multiple gray nodules surrounding the testis and epididymis, attached to the tunica albuginea and vaginalis, had smooth surface, partly whorled cut surface. Histologically, the nodules were well circumscribed, consisting of fibrous tissue, with infiltration by plasma cells and mononuclear inflammatory cells, giving the diagnosis of plasma cell granulomas.

Individual-specific variation of gene expression in peripheral blood leukocytes

Project description:DNA microarray technology is used to determine gene expression profiles of various cell types, especially abnormal cells, such as cancer. By contrast, relatively little attention has been given to expression profiling of normal tissues. Here we describe studies of gene expression in peripheral blood leukocytes (PBL) from normal individuals sampled multiple times over periods ranging from several weeks up to 6 months. We demonstrate stable patterns of gene expression that differ between individuals. Among the genes whose expression varies by individual is a group of genes responsive to interferon stimulation. Certain individuals ( approximately 10-20% of those tested) showed higher baseline levels and lower inducibility of these genes in response to in vitro interferon stimulation. These studies demonstrate the feasibility of using DNA microarrays to measure the variations in gene expression of PBL from different individuals in response to environmental and genetic factors.
