Cortical gene expression of WT, Grin1 knockdown, and Grin1 Cre rescue mice

ABSTRACT: The purpose of the study was to compare cortical transcript abundance between WT mice and Grin1 knockdown mice, and to determine whether transcript levels could be normalized in Grin1 Cre rescue mice (hemizygous for tamoxifen-inducible Rosa26 ERT2 Cre). All mice were treated with tamoxifen at 10 weeks of age until 12 weeks of age, and were euthanized at 15 weeks of age.

Project description:A comparative analysis of gene expression of 3 different experiments; 1. Perinate or adult-tagged GFP+YFP+ and bulk GFP+YFP- Tregs, 2. FL or BM-derived Tregs 3. Perinate or adult thymic Tregs. 1. Foxp3-eGFP-Cre-ERT2 x Rosa26-YFP lineage-reporter mice were injected ip with tamoxifen (4 injections, every third day) between 0-10 (perinate) or 35-45 (adult) days of age, and keep until 8 weeks of age. Perinate or adult-tagged GFP+YFP+ cells and bulk GFP+YFP- were double-sorted. 2. Hematopoietic progenitor cells were isolated from E18.5 fetal liver of CD45.1 congenic mice or from bone marrow of 5 weeks old CD45.2 B6 mice via negative selection of Thy1.2+ cells. The two populations were mixed at a 1:1 ratio, and were iv-transferred into irradiated (1000 rad) Rag-1-KO mice. FL or BM-derived Tregs were double sorted after reconstitution. 3. Foxp3-GFP+ Tregs were double-sorted from perinate (4 days old) or adult (5 weeks old) thymi. RNA from whole samples was amplified, labeled, and hybridized to Affymetrix Mouse Gene 1.0 ST Arrays.

Comparison of Hepatocyte nuclear factor 4 alpha developmental and temporal knockout mice

Project description:The goal of this experiment was to distinguish those genes regulated following acute HNF4alpha depletion compared to a developmental knockout model where gene compensation may comfound results. Expression profile of livers from 8 week old male Hnf4alpha Flox mice that express either albumin cre or the tamoxifen inducible ErT2-albumin cre for liver-specific deletion. Mice were fed a control diet or tamoxifen diet in the case of the ErT2-albumin cre to induce recombination.

Impact of IRP deficiency on the proteome of Ly6G+ cells from the bone marrow in mice

Project description:The overall objective of the project is to evaluate the effects of loss of function of iron regulatory proteins (IRP1, encoded by Aco1)-1 and -2 (IRP2, encoded by Ireb2) during adult life. Mice carrying floxed Aco1 and Ireb2 alleles were crossed to a knockin strain expressing a tamoxifen-inducible CRE recombinase under the control of the Rosa26 promoter. The resulting Aco1flox/flox,Ireb2flox/flox,Rosa26+/CreERT2 mice are designated P1/2-KO; littermates lacking the CreER sequence (Aco1flox/flox,Ireb2flox/flox,Rosa26+/+) serve as reference and are designated P1/2-CTR. Adult mice were injected (i.p.) with tamoxifen on day 1 and day 3 to ablate the IRPs in the whole body, and were sacrificed on day 10 for analysis. Bone marrow cells were collected and LY6G+ cells were magnetically sorted for proteome analysis by LC-MS/MS using an Ultimate 3000 UPLC system connected to an Orbitrap Exploris 480 mass spectrometer.

Mouse retina from Msi1/Msi2 double knockout in photoreceptor cells

Project description:This quantitative protepomics study (TMT10 isobaric labeling) of the protein expression in the retina of Msi1/Msi2 double knockout mouse compared to floxed controls. The Msi1 and Msi2 genes were knocked out in photoreceptor cells using tamoxifen inducible Cre (Cre-ERT2) under the control of Pde6g promoter. Tamoxifen was administered by intrapertoneal injection for three consecutive day starting at postnatald day 30, and the retina was collected at postnatal day 51. Retinas from floxed animals lacking the Cre recombinase and treated with tamoxifen were used as controls. Five biological replicates for each knockout (three femailes, two males) and control (two femaes and three males) group were used.

Expression profile of hematopoietic stem and progenitor cells (HSPCs) after conditional deletion of the histone 3 lysine 4 (H3K4) methyltransferase Setd1b in mice

Project description:Loss of Setd1b in adult mice using both ubiquitous and hematopoietic-specific deletion strategies caused lethality arising from severe hematopoietic defects. To identify genes regulated by Setd1b in HSPCs we performed RNA-seq analysis of the LSK (Lin-, Sca+, Kit+) compartment in the bone marrow. Mice carrying heterozygous and homozygous conditional Setd1b alleles and Rosa26-Cre-ERT2 were treated with Tamoxifen for 6 days (4 days gavage - 3 days pause - 2 days gavage). Mice were sacrificed 3 days after the last treatment and RNA was isolated and processed for next generation sequencing. We found that Setd1b regulates key lineage specification components including Cebpa, Gata-1 and Klf-1.
