Temperature induced changes in the Arabidopsis transcriptome

Project description:Plants developed plasticity to environmental conditions, such as temperature, that allows their adaptation. A change in ambient temperature leads to changes in the transcriptome in plants. Here we study changes in the splicing of Arabidopsis thaliana using an RNA-seq approach. Plants were growing in short day conditions (8h light/16h dark) at 16°C for 5 weeks and then moved to 25°C. We monitor changes in the transcriptome in the time, material was harvested 24h, 3 days and 5 days after the temperature change.

Effect of high temperature on Arabidopsis thaliana root tips

Project description:Plant roots located in the upper soil layers are prone to experience high temperatures. To gain insight into the effect of high temperature on root development and functioning, we exposed five-day-old Arabidopsis thaliana seedlings grown on agar plates to 30 °C for 48 hours, and compared the gene expression profile in the root tip with that from seedlings that remained at 22 °C.

Role of cis-zeatin in root responses to phosphate starvation

Project description:In this work we show that root illumination influences Pi starvation responses, enhancing the root and shoot growth arrest and limiting the root/shoot ratio as well as root hair elongation. A comparative transcriptomic study using dark-grown roots (DGR) roots seedlings taht were grown with or without phosphate identifies several genes that respond to Pi deficiency that were not previously reported, likely by the negative effect of the root illumination.

Metabolomics of Diapause in Aedes albopictus

Project description:Replicate populations of Aedes albopictus were reared under diapause-inducing short day photoperiod (8h light: 16h dark) and diapause-averting long day photoperiod (16h light:8h dark). Eggs were collected from each replicate and snap-frozen 11d post-oviposition. We hope to characterize the metabolomic and lipidomic profiles of diapausing eggs relative to non-diapause eggs.

Hormone profiling revealed that Pi starvation reduces the level of trans-Zeatin (tZ) but significantly increases the amount of cis-Zeatin (cZ) and cis-Zeatin Riboside (cZR). We have analyzed the transcriptomic response of Arabidopsis roots to phosphate starvation combined with cis- or trans-zeatin treatments for 8 hours.

Project description:We reported the temperature responses in roots affect the gene expression. To identify the transcriptome profiles response to temperature, we performed an mRNA-seq experiments in Col-0, hy5-215 and chl1.5 with 2 different temperature conditions (21 and 28C). The differential expressed genes in response to temperature were analyzed. It has 36 samples in total, with 3 biogical replicates for each condition.

Control of root thermomorphogenesis by systemic and local regulatory signals (18h RNA-seq)

Project description:Temperature is one of the most impactful environmental factors in response to which plants adjust their growth and development. While the regulation of temperature signaling has been extensively investigated for the aerial part of plants, much less is known and understood about how roots sense and modulate their growth in response to fluctuating temperatures. Here we found that shoot and root growth responses to high ambient temperature are negatively correlated during early seedling development. A shoot signaling module that includes HY5, the phytochromes and the PIFs exerts a central function in coupling these responses and triggers long-distance signaling to control root growth and auxin levels in the root. In addition to the HY5/PIF dependent shoot module, a local regulatory axis composed of auxin biosynthesis and auxin perception factors mediates root responses to high ambient temperature. Together, our findings uncover that roots integrate long-distance signals with local hormonal inputs during thermomorphogenesis.
