Project description:The O-glycosyltransferase LARGE2 attaches a laminin-binding matriglycan structure on alpha-Dystroglycan (α-DG) to mediate the interaction of different cell types with their laminin-containing basement membrane. We used HT-29 CRC cells, characterized by low endogenous LARGE2 gene expression, to ectopically express LARGE2 and to drive O glycosylation of its protein substrates. Immunoblot analyses had revealed that the addition of matriglycan-structures on LARGE2 substrate proteins substantially increased their molecular weight (MW). In order to take into account this shift in MW of candidate proteins, glycoprotein-enriched fractions from HT-29 cells with or without ectopic LARGE2 expression were separated by gel electrophoresis and subsequently fractionated in 6 different molecular weight windows (kDa: 0-25, 25-55, 55-75, 75-110, 110-160, 160<). Subsequently, samples were prepared for quantitative mass spectrometry analysis (qLC-MS/MS) analysis according to standard procedures. qLC-MS-MS analysis identified α-DG–related peptides within higher MW protein fractions only when LARGE2 was overexpressed. HT-29 cell secreted Laminin-2 was exclusively detected together with these high MW forms of α-DG, which demonstrated the affinity of Laminin towards the LARGE2-synthesized matriglycan structure on α-DG. Additional candidate proteins identified in this study need further experimental confirmation. Overall, we show that size fractionation after gel electrophoresis followed by quantitative mass spectrometry, performed separately on different MW windows, represents an elegant approach to reveal potential substrates of the O-glycosyltransferase LARGE2.

Project description:Low intracellular folate levels diminish the growth rate of HT-29 human colon cancer cells. This is accompanied by a metabolic shift from cytosolic glycolysis towards mitochondrial oxidative phosphorylation, as demonstrated by a lower lactate production and an increased mitochondrial oxygen consumption rate. To obtain insight in the molecular effects underlying these changes, the steady state gene expression profiles of HT-29 cells with different intracellular folate concentrations were compared. The gene expression profile of HT-29 cells with low intracellular folate levels (grown for 3 weeks in 10 ng/ml folic acid (PGA)) was clearly distinct from that of the other exposure conditions, which provide sufficient intracellular folate levels (100 ng/ml PGA, 10 ng/ml methyltetrahydrofolate (MTHF) or 100 ng/ml MTHF). Intracellular folate deficiency, contrary to expectation, did not lead to major changes in expression of genes involved in energy metabolism. This suggests that the shift towards mitochondrial oxidative phosphorylation is not mediated at the transcription level. Furthermore, only minor changes in the expression of folate metabolism related genes were observed. The changes that were observed were consistent with nucleotide salvage and in agreement with nucleotide need of the slow-growing folate-deficient HT-29 cells. The major observed effects were on cell cycle related gene expression, which was increased and interferon-responsive gene expression, which was reduced. The increase in cell cycle related gene expression seems compensatory to the reduced cell growth. Down-regulation of the interferon-response may be explained by decreased expression of signal transducer and activator of transcription 1 upon folate deficiency. We used gene microarray to analyze gene expression in HT-29, a colon cancer cell line, grown as monolayer and spheroid and identify metabolic pathways that are upregulated. HT-29 monolayer cells were grown in DMEM/F12 media supplemented with 10% FBS, and 1X Antibiotic-Antimycotic liquid (AA); and spheroids were grown in stem cell media [DMEM:F12, 1X AA, 1X B27, epidermal growth factors, and fibroblast growth factor for five days. Total RNA was isolated using the mirVana™ miRNA Isolation Kit according to the manufacturer's protocol. RNA intergrity was confirmed on the Nanodrop ND-1000 and analyzed with Agilent 2100 bioanalyzer. The arrays were scanned with the Affymetrix 3000 7G plus scanner.
