Project description:This study sought to interrogate the effects of lipids and lipid metabolites on the hepatic proteome. Protein expression in high-fat diet (HFD) mouse livers vs. livers of normal chow fed (NC) mice were investigated using multiplexed quantitative LC-MS/MS (TMT labeling). This experiment contains additional replicates for normal chow and mice on high-fat diet for 16 weeks.

Project description:Lean male mice were fed a high fat diet (HFD, lard 24% w/w) for 16 weeks. At 9 weeks, when all hallmarks of prediabetes were established, groups of mice were treated with drug (rosiglitazone, pioglitazone, T0901317, or salicylate) for another 7 weeks together with the high fat diet. An additional group was switched back to a chow diet (dietary lifestyle intervention) after the first 9 weeks of high fat diet. All groups were compared to a control group receiving HFD alone and to a reference group fed chow (baseline reference) for the entire experimental period (16 weeks).

Rat Liver HFD vs Chow, HFD vs PKCepsilon ASO

Project description:Insulin resistance drives the development of type 2 diabetes (T2D). In liver, diacylglycerol (DAG) is a key mediator of lipid-induced insulin resistance. DAG activates protein kinase C epsilon (PKCÎµ), which phosphorylates and inhibits the insulin receptor. In rats, a 3-day high fat diet produces hepatic insulin resistance through this mechanism, and knockdown of hepatic PKCÎµ protects against high fat diet-induced hepatic insulin resistance. Here we employ a systems level approach to uncover additional signaling pathways involved in high fat diet-induced hepatic insulin resistance. We used quantitative phosphoproteomics to map global in vivo changes in hepatic protein phosphorylation in chow-fed, high fat-fed, and high fat-fed with PKCÎµ knockdown rats to distinguish the impact of lipid- and PKCÎµ-induced protein phosphorylation.

Systems analysis of hepatic insulin resistance identifies RGS4 as a key mediator of metabolic adaptation

Project description:We profiled the transcriptomes of chow diet-fed and high-fat diet-fed mice across insulin stimulation time points. We also profiled the effects of metformin treatment on mouse liver transcritomes. Our temporal analyses of chow-diet and 16 week high-fat diet transcriptomics following insulin stimulation uncovered several regulator of G-protein signaling genes, particularly Rgs4, that are specifically regulated transcriptionally in HFD livers following insulin stimulation. We validated these findings with additional assays of RGS4 mRNA and protein levels and characterized this molecule's role in regulating hepatic insulin responses in mice.

Project description:The aim of this study was to characterize the obesity-related gene expression profiles between bone marrow adipocytes and peripheral white adipocytes from obese mice fed with high fat diet and leptin deficient mice Alterations of gene expression with high fat diet and in mice lacking leptin were analyzed in bone marrow and peripheral white adipocytes isolated from C57BL/6J male mice using Affymetrix Mouse Gene 1.0 ST arrays. Bone marrow adipocytes and peripheral white adipocytes (n=6-10 animals per group) were isolated from male C57BL/6J mice (6-months, 14-months ) fed with either standard chow or a high fat diet containg 60% calories from fat. Samples were grouped into diet (standard chow vs. high fat diet) and age (6-month (6M), 14-month (14M) and 18-month (18M)).

Project description:We isolated liver sinusoidal endothelial cells (LSEC) from mice fed control chow (Chow) or the nonalcoholic steatohepatitis (NASH)-inducing high fat, fructose and cholesterol (FFC) diet, and subjected the cells to bulk RNA-seq. Out of 13993 transcripts detected, the adhesion molecule Vcam1 was significantly upregulated in LSEC from FFC-fed mice compared to Chow-fed mice (Log2FC: 0.925, p: 3.6x10E-18), whereas the upregulation of another adhesion molecule Icam1 (Log2FC: 0.416, p: 0.014) was not as striking as Vcam1.
