Expression data from different tissues in different seasons from P. tomentosa

Project description:Different fuctional genes have different expression levels in various tissues. Along with the seasons changing, expression level also changes. One-year expression level for functional genes can be indicated by the four seasonal samples and respective time-course change can also be detected. We use microarry chips to study the expression levels for specific genes and explore new functional genes involved in secondary metabolism Samples from four tissues (bud, root, xylem and phloem) were collected in different seasons in one year. April and May were the months in spring for collecting samples. June and July were the months in summer for collecting samples. September and October were the months in autumn for collecting samples. December was the month for collecting samples in winter. RNA from each sample was extracted for hybridization.

Expression analysis in pancreatic and liver tissues of diabetic db/db mice in response to dietary supplementation with selenate

Project description:Investigation of gene expression level changes in pancreatic and liver tissues of diabetic db/db mice supplemented with selenate, compared to the diabetic db/db mice administered placebo. Fasting blood glucose levels increased continuously in diabetic db/db mice administered placebo (DMCtrl) but decreased gradually in selenate-supplemented diabetic db/db mice (DMSe) and approached normal values when the experiment ended. The size of pancreatic islets increased, causing the plasma insulin concentration to double in DMSe mice compared with that in DMCtrl mice. Two six chip studies using total RNA respectively isolated from pancreatic and liver tissues of three selenate-supplemented diabetic db/db mice, and three diabetic db/db mice administered placebo.

A transcriptomic resource for Douglas-fir seed development and analysis of transcription during late megagametophyte development

Project description:We developed a transcriptome resource for Douglas-fir covering key developmental stages of megagametophytes over time: prefertilization, fertilization, embryogenesis, and early, unfertilized abortion. Extracted RNA was sequenced using large-scale sequencing and reads were assembled to generate a de novo reference transcriptome of 105,505 predicted high-confidence transcripts. Expression levels were estimated based on alignment of the original reads to the reference. 200â400 megagametophytes were dissected and pooled per sample on four dates from either pollinated or unpollinated cones: June 10, June 22, June 30, and July 6 2011. These dates coincided with key events in seed development: corrosion cavity formation, fertilization, embryogenesis, or the early stages of abortion in the unpollinated treatment. Sporophytic tissue (i.e. cone bracts and cone scales) were added for comparison. PolyA RNA was used for Illumina sequencing.

circRNAs expression in the plasma of DM patients, DKD patients and healthy controls

Project description:In order to explore the role of circRNA in diabetic kidney disease (DKD), we employed circRNAs microarray expression profile in the plasma of daibetes patients (n=6), diabetic kidney disease patients (n=6) and healthy controls (n=6). According to the microarray data, we found 2079 differentially expressed circRNA in the plasma of DKD patients compared to the healthy people, including 1182 up-regulated and 897 down-regulated.Among the differentially expressed circRNA, nine DKD-related circRNAs were chosen and further validated by qRT-PCR.The results revealed that circUBXN7 was significantly elevated in the plasma of DKD patients, and might be a diagnostic plasma marker for DKD.
