GNPS - Ginseng alleviated the fatigue-induced metabolic disorder

ABSTRACT: This study used UPLC-QTOF-MS/MS based metabolomics to investigate the influence of ginseng water extracts on endogenous metabolism of fatigue rats. The results found that ginseng water extracts obviously restored the fatigue-induced bile acid, amino acid, fatty acid and lipid metabolic disorder.

Project description:The aim of the study was to investigate the involvement of the mTOR pathway in the autism spectrum disorder. Adolescent male rats, prenatally exposed to valproic acid (500 mg/kg ip on gestational day 12.5), were treated with the mTOR inhibitor rapamycin (10 mg/kg, ip, two consecutive days starting at postnatal day 35). Transcriptomic analysis was performed on the ventral portion of coronal sections, containing the striatum. Behavioral experiments, the electrophysiology on striatal slices, a morphological analysis by confocal microscopy and the determination synaptic proteins expression in the nucleus accumbens were also performed.

Thyroid Dysfunction, Neurological Disorder and Immunosuppression as the Consequences of Long-term Combined Stress

Project description:Stress is a powerful modulator of neuroendocrine, behavioral, and immunological functions. So far, the molecular mechanisms of response to stressors still remain elusive. In the current study, after 10 days of repeated chronic stress (hot-dry environment and electric foot-shock), a murine model of combinedstress (CS) was created in the SPF Wistar rats. Meanwhile, we established an ulcerative-colitis (UC) rat model induced by 2,4,6-trinitrobenzene sulfonic acid (TNBS)/ethanol enema according to previous studies. The blood, hypothalamus, and colon tissues of these rats from CS, normal control (NC), UC and sham (SH) groups, were collected for further investigations. Comparing to the NC group, the serum levels of T3, T4, fT3 and fT4 were obviously decreased in the CS group after chronic stress, indicating that thyroid dysfunction was induced by long-term combined stress. Moreover, the application of RNAseq and subsequent analyses revealed that neurological disorder and immunosuppression were also caused in the hypothalamus and colon tissues, respectively. Comparing with SH group, besides the induced colon infammation, thyroid dysfuntion and neurological disorder were also produced in the UC group, suggesting that hypothalamic-pituitary-thyroid (HPT) axis and gastrointestinal system might not function in isolation, but rather, have intricate crosstalks.

Analysis of Panax ginseng secreted proteins under salt stress

Project description:Salt stress is one of the major abiotic stresses affecting the yield of ginseng (Panax ginseng C. A. Meyer). The objective of this study was to identify proteins of ginseng, which is responsive in salt stress. In this direction, ginseng plants of different growth stages (3, 4 and 5 years), were grown in the hydroponic conditions and exposed to 5 ds/m salt concentration. The secreted proteins, collected from the water, at 0, 24, 72 and 120 hours after exposure were used for the proteome analysis using shotgun approaches. Through the shotgun proteomics, a total of 155 and 88 secreted proteins were identified by searching in two RNA-sequencing (RNA-seq) database, respectively.

Label-free Quantitative Proteomic analysis of drought-responsive proteins in Panax ginseng Meyer

Project description:Korean ginseng (Panax ginseng Meyer) has long been cultivated as an important medicinal plant. Drought results from the moderate water loss, which primarily impairs the growth of ginseng and reduction of yield loss. However, basis of biological clues to understanding the accurate mechanisms related to drought stress in proteome level are still elusive. Therefore, we carried out label-free quantitative proteomic analysis using ginseng roots subjected to drought stress which was grown at less than 10% soil moisture for two weeks, compared with normal ginseng which was grown at 25% soil moisture. The acquired proteins were carried out label-free proteomic analysis using LC-MS/MS. This approach led to the identification of total 2,471 proteins, and out of 195 proteins showed significant modulation. Functional classification revealed that proteins related to secondary metabolites, calcium signaling, and photosynthesis were enriched in control sample (cluster_1), while proteins associated with stress responsive, redox, electron transport, and protein synthesis were mainly dominated in cluster_2 (drought stress condition). Taken together, our results provided an overview of the drought-induced proteomic changes in ginseng root, and it is correlated with physiological changes, contributing to reveal potential marker at proteome level in ginseng.

A systems-level analysis of bile acids effects on rat colon epithelial cells

Project description:Bile acid diarrhoea is a chronic condition caused by increased delivery of bile acids to the colon. The underlying mechanisms remain to be elucidated. To investigate genes involved in bile acid diarrhoea, systems-level analyses were employed on a rat bile acid diarrhoea model. Twelve male Wistar Munich rats, housed in metabolic cages, were fed either control or bile acid-mixed (1% w/w) diets for ten days. Food intake, water intake, urine volume, bodyweight and faecal output were monitored daily. After euthanasia, colonic epithelial cells were isolated using calcium-chelation and processed for systems-level analyses, i.e. RNA-sequencing transcriptomics and mass spectrometry proteomics. Bile acid-fed rats suffered diarrhoea, indicated by increased drinking, faeces weight and faecal water content compared with control rats. Urine output was unchanged. With bile acid-feeding, RNA-sequencing revealed 204 increased and 401 decreased mRNAs; mass spectrometry 183 increased and 111 decreased proteins. Among the altered genes were genes associated with electrolyte and water transport (including Slc12a7, Clca4 and Aqp3) and genes associated with bile acid transport (Slc2b1, Abcg2, Slc51a, Slc51b and Fabps). Correlation analysis showed a significant positive correlation (Pearson’s r=0.28) between changes in mRNA-expression and changes in protein-expression. However, caution must be exercised in making a direct correlation between experimentally determined transcriptomes and proteomes. Genes associated with bile acid transport responded to bile acid-feeding, suggesting that colonic bile acid transport occur by regulated protein facilitated mechanisms rather than passive diffusion. In addition, the study provides annotated rat colonic epithelial cell transcriptome and proteome with response to bile acid-feeding.

Bipolar disorder and lithium-induced gene expression in two peripheral cell models

Project description:Bipolar disorder is a severe and heritable psychiatric disorder and affects up to 1% of the population worldwide. Lithium is recommended as first-line treatment for the maintenance treatment of bipolar-affective disorder in current guidelines, its molecular modes of action are however poorly understood. Cell models derived from bipolar patients could prove useful to gain more insight in the molecular mechanisms of bipolar disorder and the common pharmacological treatments. As primary neuronal cell lines cannot be easily derived from patients, peripheral cell models should be evaluated in their usefulness to study pathomechanisms and the mode of action of medication as well as in regard to develop biomarkers for diagnosis and treatment response.
