Novel inducible filamentous sliding motility identified in pathogenic Bacillus cereus group species

Project description:We isolated an atmospheric contaminant, subsequently identified as a new strain of Bacillus mobilis, which showed a novel, robust, inducible filamentous sliding motility and completely colonized a bacterial culture plate in less than 48 h under some conditions. This flagella-independent sliding motility was characterized by long filamentous cells at the expanding edge, and was induced when cells were inoculated onto lawns of metabolically inactive Campylobacter jejuni cells, heat killed bacterial biomass, and milk or blood dried onto agar plates. Phosphatidylcholine (PC), bacterial membrane components, and sterile human fecal extracts were sufficient to induce filamentous expansion. Screening of eight other Bacillus spp. (five from the B. cereus group and three other Bacillus spp.) showed that filamentous motility was conserved amongst B. cereus group species to varying degrees. RNAseq of filamentously expanding cells collected from PC and milk lawn plates in comparison to rod-shaped cells from control plates revealed that genes related to metabolism, ion and amino acid transport were differently regulated, genes controlling sporulation were reduced, and some virulence genes (e.g., hblA/B/C/D and plcR) were increased. We hypothesize that the robust and conserved nature of filamentous motility in pathogenic B. cereus group species can enhance bacterial colonization during host colonization.

Large-scale differences in gene expression, including genes encoding type IV pili-associated proteins, are seen when Clostridium perfringens is grown in liquid or on surfaces

Project description:Clostridium perfringens is a Gram-positive anaerobic pathogen that causes multiple diseases in humans and animals. C. perfringens lack flagella but have type IV pili (TFP) and can glide on agar surfaces by forming filaments of cells aligned end to end. When cells are placed on agar surfaces, they become elongated, flexible and have TFP on their surface. To understand the basis of these phenotypes, cells were grown in three types of liquid media and on agar plates with the same medium to compare gene expression using RNA-seq. Hundreds of genes were differentially expressed, including genes associated with TFP functions, which were higher on plates than in liquid. The gusA gene, encoding the enzyme β-glucuronidase, was inserted into the chromosome downstream of TFP promoters and expression measured in liquid and on plates. β-glucuronidase activity was proportional to the amount of transcripts seen with RNA-seq n liquid-grown cells, but not plate-grown cells, suggesting post-transcriptional regulation of these genes on surfaces. An sRNA adjacent to pilA1 (SR79) was expressed at higher levels on plates in all media; overexpression of this sRNA resulted in longer cells on plates and shorter lag times in liquid suggesting SR79 plays a role in adapting to surface growth.

Project description:Dataset 1 contains bacterial-fungal monocultures and tripartite co-cultures in which Scopulariopsis sp. JB370 are grown with 1) Hafnia alvei JB232 and 2) Pseudomonas psychrophila JB418 or Escherichia coli K12. Dataset 2 contains bacterial-fungal tripartite co-cultures in which Penicillium solitum #12 are grown with 1) Glutamicibacter arilaitensis JB182 or Brevibacterium linens JB5 and 2) Pseudomonas psychrophila JB418 or Escherichia coli K12. All cultures from each dataset were grown on 10% cheese curd agar (CCA) and extracted using acetonitrile. Extracts were analyzed via LC-MS(/MS) and processed with MZmine2 for analysis with MetaboAnalyst 5.0 and GNPS feature based molecular networking.

Beauveria bassiana SNF 1 (BbSNF1) regulating genes involved in conidiation

Project description:Gene BbSNF1 playes an essential role in mediating metabolism and cell development during conidation, a determinant to the biocontrol potential of entomopathogenic fungi. The genome-wide exprssion analysis involved in fungal conidiation was analyzed by using high throughput sequencing (RNA-Seq). Our transcriptional profiles revealed that numerous differentially expressed genes (DEGs), of which involved in metabolism, cell transport and cell rescue, were significantly involved in deveopment of fungal conida. . Total RNA obtained from BbSNF1 disruption mutant were compared to that of wild type strain (WT) grown on SDA plates (Glucose 4%, Peptone 1%, and Yeast extract 1% plus 1.5% Agar).
