Project description:Dendritic cells (DC) are the most important antigen presenting cells and play a pivotal role in host immunity to infectious agents by acting as a bridge between the innate and adaptive immune systems. Myeloid DCs (mDC) were isolated from peripheral blood and co-cultured with conidia (con) and germlings (ks) of Aspergillus fumigatus knockout (mitA, mnt1, rodA) and wildtype (wt) strains for 6 hours at an MOI of 1. RNA was extracted and hybridised to microarrays. Microarrays contain probes for approximately 110 selected human immune-relevant genes, including cytokine and chemokine genes, their receptors and downstream innate immunity genes. We used microarrays to detail the gene expression of human myeloid dendritic cells after 6h of co-cultivation with Aspergillus fumigatus wildtype and knockout mutants

sRNA-seq of Aspergillus fumigatus RNAi double knockouts

Project description:The RNA interference (RNAi) pathway has evolved numerous functionalities in eukaryotes, with many on display in Kingdom Fungi. RNAi can regulate gene expression, facilitate drug resistance, or even be altogether lost to improve virulence potential in some fungal pathogens. In the WHO fungal priority pathogen, Aspergillus fumigatus, the RNAi system is known to be intact and functional. To extend our limited understanding of A. fumigatus RNAi, we performed a multi-condition sRNA-seq analysis comparing expression of several RNAi double knockout mutants with the wild-type strain in conidia and mycelium grown for 24 or 48 hours.

Proteome of three strains of Aspergillus on tea leaves

Project description:Aspergillus were believed the dominant fungi in Post-fermentation Pu-erh tea (Pu-erh Shucha, PFPT), a well-known traditional Chinese tea with multiple healthful benefits. However, the functions of Aspergillus in PFPT fermentation lack deeply investigated. In this work, A. niger, A. tamarii and A. fumigatus were fermented on sun-dried green tea leaves, and these samples (marked as Asn, Ast and Asf) were analyzed by integration of proteomics and metabolomics methods. We identified 2035, 2210 and 1095 Unique proteins in Asn, Asf and Ast, and assigned majority of identified proteins to cellular process and metabolic process in GeneOntology (GO) analysis. Majority KEGG pathways enriched significantly belonged to Metabolism. Through dbCAN annotation, 232, 205 and 226 proteins were annotated to 103, 86 and 97 families or subfamilies of Carbohydrate-active enzymes (CAZymes) in Asn, Asf and Ast, respectively. CAZymes involved in degradation of cellulose, starch, lignin, pectin, xylan and xyloglucan were identified, such as endoglucanases, glucosidase, cellulase. We further identified enzymes hypothesized to hydrolysis, oxidization, polymerization, conversion, biosynthesis and degradation of polyphenols or benzenoids. Together, we found that Aspergillus. spp produced enzymes in tea leaves, which involved in the degradation of plant polysaccharides and metabolism of tea polyphenols.

Project description:Deletion of transcription factors DevR and AcdX in Aspergillus fumigatus causes increased/decreased susceptibility to the antifungal Olorofim. In order to characterise the basis of differential Olorofim susceptibility, and distinguish which metabolic pathways these transcription factors regulate, RNA-seq analysis was used to compare these transcription factor knockout mutants to wildtype A. fumigatus in both the presence and absense of Olorofim.

Expression data from batch cultivations of Aspergillus niger wild type and adrA, facB and creA deletion mutants

Project description:Expression data from batch cultivations of Aspergillus niger wild type strain ATCC 1015 and adrA, facB and creA deletion mutants constructed on ATCC 1015 background strain with glucose or glycerol as carbon sources. Genome-wide transcriptome analysis was used to identify genes either affected directly or indirectly by each transcription factor investigated during growth on a repressing or a derepressing carbon source. For this purpose, batch cultivations under well-controlled conditions were performed with Aspergillus niger wild type strain ATCC 1015 and the three deletion mutants of the corresponding transcription factors AdrA, FacB and CreA. Samples for RNA extraction were collected and further processed for hybridization in custom-designed Affymetrix microarrays containing probes for three Aspergillus species, including A. niger.
