Transcription profiling of wild type and RAP2.2 over-expressing Arabidopsis leaves

Project description:Comparison of rosette leaves of two different RAP2.2 overexpressing lines with wild type leaves. The AP2/EREBP transcription factor RAP2.2 was shown to bind to a cis-acting motif within the phytoene synthase promoter from Arabidopsis. To investigate effects of increased RAP2.2 levels, two RAP2.2 overexpressing Arabidopsis thaliana (ecotype Wassilewskija) lines were generated: one line, nosr2, carried the nos promoter and showed a two-fold increase in RAP2.2 transcript level, the second line, cmr-5, carried four copies of the CaMV-35S enhancer and showed a 12-fold increase. However, neither weak nor strong increase in RAP2.2 transcript amounts had any effect on RAP2.2 protein levels as shown by Western blot analysis. The strong robustness of RAP2.2 protein levels towards transcriptional changes can be explained by specific protein degradation which includes SINAT2, an E3 ubiquitin ligase which was isolated using a two-hybrid approach. Accordingly, global gene expression analysis using both RAP2.2 overexpressing lines showed only minor transcriptional changes which are probably due to minor growth variation than to mechanisms involved in the down-regulation of RAP2.2 protein amounts.

Targets of AtWRKY27 in Arabidopsis leaves

Project description:The Arabidopsis transcription factor WRKY27 was found to be involved in plant defense towards Ralstonia solanacearum GMI1000. To identify target genes of WRKY27, we introduced a functional tagged version of WRKY27 into two independent Arabidopsis wrky27 knockout lines (ecotype Columbia) under the control of the estrogen receptor-based chemical-inducible system. 18 days old plants grown on soil (Metro Mix 200) at 22oC under a 10/14 h light/dark cycle were treated for 6h with 10 microM beta-estradiol after which RNA was immediately isolated. The two independent knockout lines transformed with the empty vectors (pMD::XVE-SALK and pMD::XVE-ETL) served as controls and were grown under the same conditions and treated identically as the experimental plants. Project description:The focus of this study was to identify changes in host gene expression induced by the transcription-dependent function of the viral AC2 protein, and induced by the interaction of AC2/C2 with SnRK1.2 (AtAKIN11). We used microarrays to identify changes in host gene expression induced by the transcription-dependent function of the geminivirus-encoded AC2 protein, and induced by the interaction of AC2 with a host serine-threonine kinase, SnRK1. Distinct classes of genes were up- and down regulated during this process. For experiment 1, Arabidopsis rosette leaves were infused with Agrobacterium capable of expressing full-length Cabbage leaf curl virus (CaLCuV) AC2 protein, a truncated version of AC2 lacking the C-terminal 29 amino acids (AC2del), or an empty vector control (530). In experiment 2, Arabidopsis rosette leaves were infused with Agrobacterium capable of expressing full-length Spinach curly top virus C2 protein, a truncated version of C2 lacking the C-terminal 29 amino acids (C2del), antisense (as) SnRK1.2 (AsAKIN11), or an empty vector control (530).

Project description:Flavonols are a class of flavonoids. Unlike other classes of flavonoids, flavonols strongly accumulate in guard cells, but the role of their accumulation in guard cells is still unclear so far.The overexpression of TaFLS1, a flavonol synthase gene from wheat, improves flavonol content in guard cells of Arabidopsis, while the mutant fls1-3 of Arabidopsis AtFLS1 reduces the content. Both the increase of flavonols by TaFLS1 overexpression and the decrease in fls1-3 change stomatal apeture, showing the association between flavonols and stomatal movements. We used microarrays to detail the global programme of gene expression underlying both the increase and decrease of flavonol content and identified possible mechanism of flavonols-mediated stomatal movement. Arabidipsis Col-0, TaFLS1 overexpression lines, and fls1-3 were used for analysis. Leaves of six-week old Arabidopsis seedlings grown in soils under the regime of a 16 h photoperiod at 22°C and 60% relative humidity were sampled, and RNA was extracted for microarray analysis. Each of Arabidopsis lines has three replicates.
