Drosophila gene expression in response to the botanical insecticide Piper nigrum

Project description:This experiment was used to determine the effect of a botanical insecticide upon gene expression profiles in Drosophila melanogaster. Adult female Drosophila (oregon-R strain) were treated with an ethylacetate extract of Piper nigrum (Piperaceae) seeds formulated in 99% ethanol. Treatment was topical, using a Potter's tower to administer a total of 2 mL of a 0.9mg/mL concentration. Control treatment was identical except flies were treated with 99% ethanol as a solvent control. Gene expression was studied four hours post-treatment. The ‘PepSAVI-MS’ pipeline for natural product bioactive peptide discovery

Project description:The recent increase in extensively drug resistant bacterial pathogens and the associated increase of morbidity and mortality demonstrate the immediate need for new antibiotic backbones with novel mechanisms of action. Here, we report the development of the PepSAVI-MS pipeline for bioactive peptide discovery. This highly versatile platform employs bioinformatics, statistics, and mass spectrometry to identify bioactive peptide targets from complex biological samples. We validate the use of this platform through the successful identification of known bioactive peptides from a botanical species, Viola odorata. Using this pipeline, we have widened the known antimicrobial spectrum for V. odorata cyclotides, including antibacterial activity of cycloviolacin O2 against E. faecium and A. baumannii. We further demonstrate the broad applicability of the platform through the identification of novel anticancer activities for cycloviolacins by their cytotoxicity against ovarian, breast and prostate cancer cell lines.

A Natural Botanical Product, Resveratrol, Effectively Suppresses Vesicular Stomatitis Virus Infection In Vitro.

Project description:Numerous natural phytochemicals such as resveratrol are acknowledged as potent botanical agents in regulating immune responses. However, it is less understood whether such immunomodulatory phytochemicals are appropriate for use as direct treatments in veterinary viral diseases. In the present study, we investigated the efficacy of resveratrol in suppressing vesicular stomatitis virus (VSV) infection. Outbreaks of VSV can cause massive economic loss in poultry and livestock husbandry farming, and VSV treatment is in need of therapeutic development. We utilized a recombinant VSV that expresses green fluorescent protein (GFP) to measure viral replication in cells treated with resveratrol. Our findings revealed that resveratrol treatment affords a protective effect, shown by increased viability and reduced viral replication, as indicated by a reduction in fluorescent signals. Additionally, we found that resveratrol inhibition of VSV infection occurs via suppression of the caspase cascade. Structural analysis also indicated that resveratrol potentially interacts with the active sites of caspase-3 and -7, facilitating antiviral activity. The potential effect of resveratrol on reducing VSV infection in vitro suggests that resveratrol should be further investigated as a potential veterinary therapeutic or prophylactic agent.

Characterization of loquat (Eriobotrya japonica Lindl.) honey botanical origin according to its chitinase activity

Project description:Bees make honey from the nectar that they collect from flowers. The characteristics of honey are closely associated to original botanical species. Compare with sugars in honey, proteins are minor components but usually used as an important honey quality evaluation parameters. Flower-origin proteins could be a good marker for the authentication. However, as a minute component in honey proteome, plant origin proteins are hard to be detected in honey by regular proteomic approaches, such as gel-based techniques. In this study, Eriobotrya japonica Lindl. (loquat) nectar and its derivative monofloral honey were systematically compared, especially regarding the proteomes and enzymatic activities. Using two-dimensional electrophoresis and mass spectrometry, only bee-originated proteins were detected in loquat honey which were major royal jelly proteins and two uncharacterized proteins. Xylosidase, thaumatin, and two kinds of chitinases were detected in loquat floral nectar by the gel-based proteomic approach. To our knowledge, it is the first study to analysis nectar-originated enzymes’ activity in honey and we proposed that the zymography of chitinase is a potential marker for honey botanical origin authentication.
