Project description:The aim of the study was to carry out a CGH study utilizing a set of 39 diverse Bacillus isolates. Thirty four B. cereus and five B. anthracis strains and isolates were chosen so as to represent different lineages based on previous characterizations, including MLEE and MLST (Helgason, Okstad et al. 2000; Helgason, Tourasse et al. 2004). They represent the spectrum of B. cereus phenotypic diversity by including soil, dairy and periodontal isolates in addition to virulent B. anthracis strains.

A coordinated attack by a bacterial secretion system and a small molecule drives prey specificity

Project description:Vibrio species are recognized for their role in food- and water-borne diseases in humans, fish, and aquatic invertebrates. We screened bacterial strains isolated from raw food shrimp for those that are bactericidal to Vibrio strains. Here we identify and characterize Aeromonas dhakensis strain A603 which shows robust bactericidal activity specifically towards Vibrio and related taxa but less potency toward other Gram-negative species. Using the A603 genome and genetic analysis, we show that two antibacterial mechanisms account for its vibriocidal activity -- a highly potent Type Six Secretion System (T6SS) and biosynthesis of a vibriocidal phenazine-like small molecule, named here as Ad-Phen. Further analysis indicates coregulation between Ad-Phen and a pore-forming T6SS effector TseC, which potentiates V. cholerae to killing by Ad-Phen.

Diamide-based screening method for isolation of improved oxidative stress tolerance phenotypes in Bacillus mutant libraries

Project description:The bacterium Bacillus subtilis is of high importance both as a model organism for Gram-positive bacteria and as an industrial work horse in the production of biomolecules. In recent years, advancements have been made to engineer the bacterium even further towards industrial applications. In this study, we present a novel screening method for mutant libraries using diamide, an oxidising agent that binds free thiols and creates disulfide bonds between them, thereby causing a so-called ‘disulfide stress’ in bacteria. The method shows promise to selectively identify phenotypes in B. subtilis with improved tolerance towards oxidative and disulfide-associated stress. Phenotypes initially identified by transposon mutagenesis were recreated through targeted gene deletions. Among the resulting deletion mutants, the largest difference in diamide tolerance compared to the parental strain was observed for pfkA and ribT deletion strains. A proteomics analysis showed that that diamide tolerance can be achieved through different routes involving increased expression of stress management proteins and reduced availability or activity of the RNA degradosome. We conclude that our screening method allows the facile identification of Bacillus strains with improved oxidative stress tolerance phenotypes

New genetic biomarkers to differentiate pathogenic and clinically relevant Bacillus cereus strains

Project description:Bacillus cereus is the second leading cause of collective food poisoning in France. B. cereus is also associated with severe clinical infections leading to patient death in 10% of the cases. The emergence of B. cereus as a foodborne and opportunistic pathogen has intensified the need to distinguish strains of public health concern. In this work, by performing a screen on a large collection of B. cereus strains of varying pathogenic potential, we identified genetic determinants capable of discriminating B. cereus strains inducing negative clinical outcomes. The combination of 4 biomarkers is sufficient to accurately discern clinical strains from harmless strains. Three of the biomarkers are located on the chromosome, with a fourth one identifying a plasmid carried by most pathogenic strains. A 50 kbp region of this plasmid promotes the virulence potential of these strains and could thus be defined as a new pathogenicity island of B. cereus. These new findings help in the understanding of B. cereus pathogenic potential and complexity and may provide tools for a better assessment of the risks associated with B. cereus contamination to improve patient health and food safety.

RNA-Seq gene expression profiling in the plant growth promoting Bacillus atrophaeus UCMB-5137 revealed an alternative strategy of plant colonization compared to Bacillus amyloliquefaciens endophytes

Project description:Plant growth promoting rhizobacteria (PGPR) of the genus Bacillus are successfully used as biofertilizers and biopesticides. They potentially can reduce the use of chemicals in agriculture as an ecologically safe alternative, but to optimize the application of PGPR, more profound knowledge on specific gene regulation and molecular mechanisms of interaction with plants is needed. Advance in sequencing technologies made it affordable to compare transcriptom profiles of relative organisms to check to which extend PGPR strains or closely related species differ in their strategies of plant colonization. This work aimed at analysis of gene regulation in a biotechnological strain Bacillus atrophaeus UCMB-5137 to compare it with the gene expression profile of a generally recognized PGPR strain B. amyloliquefaciens FZB42. It was found out that despite the close taxonomic relatedness, these two organisms developed ability to colonize plants independently and use different strategies of plant colonization. Root exudate has triggered in UCMB-5137 alteration in expression in many genes controlled by stress response transcription factors (TF) SigB and SigD, while SigF, SigH, SigW, CcpA and several other TFs regulated genes associated with quorum sensing and biofilm formation, and adjusted the carbohydrate metabolism. Counting to peculiarities of gene regulation in different PGPR strains will allow optimization of their practical application.
