GNPS - Microbiome Reciprocal Transfer Experiment 2023 - Investigating the effect on the host of `mismatched` microbiomes

ABSTRACT: Full reciprocal transfer of microbiomes between nine host strains of phytoplankton and microbe-free controls (Kuijpers et al unpublished). Microbiomes were naturally recruited from lake and stream water in an assembly experiment (Broe et al unpublished). Transferred microbiomes were added to axenic hosts and grown for 7 or 14 days before control or heat shock treatment.

Quantitative proteomics of biofilm formation of the Antarctic archaeon Halorubrum lacusprofundi

Project description:Halorubrum lacusprofundi is an important member of Deep Lake in Antarctica, representing ~10% of the lake population (DeMaere et al., 2013). Deep Lake is in the Vestfold Hills, East Antarctica (68°33’36.8S, 78°11’48.7E) and is 36 m deep, monomictic, and perennially cold (down to -20°C) (DeMaere et al., 2013; Williams et al., 2014; Tschitschko et al., 2015; Tschitschko et al., 2016). In this project, using quantitative proteomics (8-plex iTRAQ labeling), proteins, pathways and cellular processes important for biofilm formation were determined for Hrr. lacusprofundi. The data provided a new level of understanding about molecular mechanisms and regulatory networks involved in adaptation of Hrr. lacusprofundi to biofilm formation. DeMaere, M. Z., Williams, T. Study for Validation of Immunological Biomarkers in Patients With Metastatic Colorectal Cancer

Project description:The investigators recently identified promiscuous HLA-DR-derived epitopes from the human telomerase reverse transcriptase (TERT) called universal cancer peptides (UCP), to study tumor-specific CD4+ T cell responses. The investigators found high frequency of naturally occuring UCP-specific TH1 cells in long term survival of metastatic colorectal cancer (CRC) previously treated by 5 fluoro-uracil -oxaliplatin (Folfox) +/- bevacizumab regiment (Godet et al. OncoImmunology 2012 and unpublished data). Epitopes-CRC02 is a French prospective multicenter study which will evaluate the post chemotherapy and post surgery modulation of host tumor-specific CD4 TH1 cell responses in metastatic colorectal cancer patients and their correlation with progression-free survival.

Genomic and transcriptomic analyses of sebacinoid fungi

Project description:The recent release of a large number of genomes from ectomycorrhizal, orchid mycorrhizal and root endophytic fungi have provided deep insight into fungal lifestyle-associated genomic adaptation. Comparative analyses of symbiotic fungal taxa showed that similar outcomes of interactions in distant related root symbioses are examples of convergent evolution. The order Sebacinales represents a sister group to the Agaricomycetes (Basidiomycota) that is comprised of ectomycorrhizal, ericoid-, orchid- mycorrhizal, root endophytic fungi and saprotrophs (Oberwinkler et al., 2013). Sebacinoid taxa are widely distributed from arctic to temperate to tropical ecosystems and are among the most common and species-rich groups of ECM, OM and endophytic fungi (Tedersoo et al., 2012, Tedersoo et al., 2010, Oberwinkler et al., 2013). The root endophyte Piriformospora indica and the orchid mycorrhizal fungus S. vermifera (MAFF 305830) are non-obligate root symbionts which were shown to be able to interact with many different experimental hosts, including the non-mycorrhizal plant Arabidopsis thaliana. These two fungi display similar colonization strategies in barley and in Arabidopsis and the ability to establish beneficial interactions with different hosts (Deshmukh et al., 2006). Colonization of the roots by P. indica and S. vermifera results in enhanced seed germination and biomass production as well as increased resistance against biotic and abiotic stresses in its experimental hosts, including various members of the Brassicaceae family, barley, Nicotiana attenuata and switchgrass (Ghimire, 2011, Ghimire et al., 2009, Ghimire et al., 2011, Waller et al., 2008, Barazani et al., 2007, Deshmukh et al., 2006). Microarray experiments were performed to identify and characterize conserved sebacinoid genes as key determinants in the Sebacinales symbioses.

Memo1 promotes FGF23-driven signaling by oxidative modulation of small Rho-GTPases and protein tyrosine phosphatase activity

Project description:Memo is a copper-dependent redox enzyme modulating receptor tyrosine kinase signaling by unknown mechanisms (Meira et al., Nat Cell Biol 2004; MacDonald et al., Science Signal 2014). Memo1 deletion causes a phenotype partially resembling Klotho and Fgf23-deficient mice (Haenzi et al., FASEB J 2014; Moor et al., JBMR Plus 2018) and increases renal abundance of Rho-GTPase RhoA but prevents FGF23-dependent renal ERK phosphorylation and Rho-GTPase Rac1 activation (unpublished). To delineate potential effects of Memo’s redox function on FGF23-driven cellular signaling processes, we performed a redox proteomics screen which identified differently oxidized Rho-GTPase chaperone protein ARHGDIA/Rho-GDP dissociation inhibitor 1 (RhoGDI) as an interaction partner of Memo. RhoGDI is regulated by cysteine oxidation and phosphorylation. Here, we investigated the potential interaction between MEMO and RhoGDI by co-incubation of recombinant proteins MEMO and RhoGDI in cell-free environment to assess the Cys79 oxidation state of RhoGDI.
