Proteomic study of aqueous humor and its application in the treatment of neovascular glaucoma

ABSTRACT: We provided a comprehensive AH proteome analysis, and expanded the profile of human AH proteome. The differential AH proteomic analysis of NVG treatment indicated that AH proteome can reflect the pathophysiological changes in drug intervention.

Aqueous humor (AH) proteins are involved in many physiological and pathological processes of the eye. The proteome analysis of AH is important to understand its physiological and pathophysiological functions. In the present study, AH samples obtained from 21 cataract volunteers were pooled together. Integrated analysis of transcriptome and proteome of the human cornea and aqueous humor reveal novel biomarkers for corneal endothelial cell dysfunction

Project description:Previous studies have reported that elevated protein levels in the aqueous humor (AH) are associated with corneal endothelial cell dysfunction (CECD), but the detailed underlying mechanism as well as specific biomarkers for CECD remain elusive. In the present study, we aimed to identify protein markers in AH directly associated with changes in Corneal endothelial cells (CECs), as AH can be easily obtained for analysis. We carried out an in-depth proteomic analysis of patient-derived AH as well as transcriptomic analysis of CECs from the same patients with bullous keratopathy (BK) resulting from CECD. We first determined differentially expressed genes (DEGs) and differentially expressed proteins (DEPs) from CECs and AH in CECD, respectively. By combining transcriptomic and proteomic analyses, 13 shared upregulated markers were observed between DEGs and DEPs. Through data integration with individual analysis via data-independent acquisition, three upregulated transcripts and proteins were identified, namely metallopeptidase inhibitor 1 (TIMP1), Fc fragment of IgG binding protein (FCGBP), and angiopoietin-related protein 7 (ANGPTL7). Furthermore, we confirmed AH biomarkers for CECD using individual proteome verification and immunoassay validation. Conclusively, our findings may provide valuable insights into the disease process and identify biofluid markers for the assessment of CEC function during BK development.

Proteomics of aqueous humor for biomarkers discovery in retinoblastoma

Project description:Background: Recent therapeutic advances have greatly improved the eye preservation rate in patients affected by retinoblastoma (RB), however local tumor control remains more difficult in the presence of resistant and/or relapsing retinal or vitreous disease. Thus, the identification of new biomarkers is crucial to design more effective treatment approaches. Recently, technological advances have allowed to safely recover aqueous humor (AH) from patients affected by RB undergoing local treatment. Methods: in order to identify specific proteins associated to active vitreous seeding, we have performed proteomic analysis of AH samples derived from patients affected by RB in different disease stages. Fifty-five AH samples were firstly included in this study. A new widened cohort was then used to validate the proteins identified in the previous discovery set. Results: a total of 808 and 630 proteins were identified in the two analysis, respectively. Five proteins resulted more expressed in the active vitreous seeding group: C19orf57, Girdin, SRFBP, GULP-1 and DCD. These proteins are involved in cytoskeleton regulation, tumor proliferation and growth, invasion and migration of cancer cells. Conclusion: reliable prognostic biomarkers are needed to guide RB management. Proteomic analysis of AH provide a unique opportunity to identify tumor associated biomarkers in patients with RB.

Project description:Background: The underlying mechanism as well as diagnostic molecular biomarkers for corneal endothelial cell dysfunction (CECD) remain elusive. This study aimed to elucidate molecular mechanism of CECD via proteomic and transcriptomic approaches and identify novel biomarkers in aqueous humor (AH) directly associated with changes in CECs. Methods: We carried out an in-depth proteomic analysis of patient-derived AH as well as transcriptomic analysis of CECs from the same patients with bullous keratopathy (BK) resulting from CECD. Selected candidate proteins were further verified by individual data-independent acquisition and immunoassay using further AH samples from CECD patients. Findings: Using integrated multi-omics analysis, systematic alterations of proteins in AH and mRNA in CECs were mainly associated with the immune mechanism during BK development. Through individual proteomic verification via DIA analysis, three AH proteins were determined as potential predictive markers including TIMP1, FCGBP, and ANGPTL7. Finally, we confirmed these biomarkers using immunoassay in individual AH samples of CECD. Interpretation: Our multi-omics analysis may provide valuable insights into the disease-specific process as well as possibility of biofluid biomarkers in CECD. Further investigation with large scale is required to determine whether AH biomarkers are useful as liquid biopsy targets for the assessment of CEC function
