Protein profile of marine gastropods

ABSTRACT: The results obtained in this study will further enrich the molecular developmental biology information of conchs and even marine bivalves. They provide important clues for elucidating the early embryonic development mechanisms of conchs and offer theoretical guidance for breakthroughs in the artificial cultivation techniques of N. arthritica cumingii.

Project description:Ophiocordyceps sinensis (Berk.) Sacc., a complex of larval carcass (sclerotium) and stroma formed by the fungus of Hirsutella sinensis infecting Hepialidae insect larvae, whose fruiting body is also the main fungal structure used for taxonomic identification. However, the induction of fruiting body is still inefficient and the high cost resulting in the large-scale artificial cultivation of this fungus has been unsuccessful in China.In this study,important factors and target genes associated with the fruiting body induction during the development of O. sinensis were identified, providing a basic molecular mechanism for facilitating the large-scale artificial cultivation of O. sinensis.

Metabolomic investigations on Nesterenkonia flava from different origins revealed significant intraspecies differences between marine and terrestrial actinomycetes

Project description:Marine is one of the most important resources of microorganisms, including bacteria, actinomycetes, and fungi. As marine and terrestrial environments differ a lot in many aspects it is not surprising that the species and characteristics of microorganisms living there are very different. Interestingly, many marine microorganisms can find their congeners of the same species from terrestrial resources. The aim of this work is to evaluate the intraspecies differences between marine and terrestrial actinomycetes on metabolic level and to uncover the mechanism responsible for the differences. To address this, we carried out comparative metabolomics study on Nesterenkonia flava strains isolated from marine and terrestrial environments. The results showed that marine strains were clearly distinguished from their terrestrial congeners on the principal components analysis (PCA) scores plot of intracellular metabolites. The markers responsible for the discrimination of marine and terrestrial strains were figured out using loading plot from partial least squares discrimination analysis (PLS-DA). Pathway analysis based on PLS-DA, univariate analysis, and correlation analysis of metabolites showed that the major differential metabolites between the terrestrial N. flava and the marine ones were involved in osmotic regulation, redox balancing, and energy metabolism. Together, these insights provide clues as to how the previous living environment of microbes affect their current metabolic performances under laboratory cultivation conditions.

Proteomic analysis of mycelial exudate of Ustilaginoidea virens

Project description:Rice false smut (RFS) is a kind of fungal disease transforming panicles and spikelets into greenish spore balls, caused by Ustilaginoidea virens. During artificial cultivation process, macroscopic exudates could be observed, which is a common feature in many kinds of fungi. We characterized the proteome of exudate associated with this plant pathogen. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis was used to identify proteins in the pathogen exudate. A total of 685 proteins comprising 3,949 peptides were identified from the exudate. This study regarded the biological characteristics of U. virens as an entry point. By studying the protein components of the exudates of U. virens, it is helpful to better understand the occurrence and pathogenic mechanism of pathogen and provide a theoretical basis for the control of RFS.

Transcriptome and metabolome analyses reveal the regulatory network governing red fruitbody disease in Morchella

Project description:True morels (Morchella spp., Morchellaceae, Ascomycota), a delicious edible mushroom, has rapidly expanded in recent years, especially in China. However, a severe disease of morels, red fruitbody disease, led to very low production of fruiting bodies. The cause reason and the mechanisms under red fruitbody are unclear. Herein, we integrated the transcriptomics and metabolomics data of M. sextelata from red fruitbody group (R) and normal group (N), which was artificial cultivation in Fujian province, China. Transcriptome data revealed the differentially expressed genes (DEGs) between R group and N group were significantly enriched in the pathways of tyrosine metabolism, riboflavin metabolism, and glycerophospholipid metabolism. Similarly, the differential accumulated metabolites (DAMs) were mainly assigned to metabolism categories, including tyrosine metabolism, biosynthesis of plant secondary metabolites, biosynthesis of amino acids, and others. Then, combined analysis of the transcriptome data and metabolome traits revealed that the most enriched pathway was tyrosine metabolism, followed by ABC transporters, alanine, aspartate and glutamate metabolism, and others. In summary, this integration of transcriptomics and metabolomics data of M. sextelata during fruitbody redness implicated several key genes, metabolites, and pathways involved in this disease. We believe that these findings will help us understand the mechanisms under fruitbody redness of M. sextelata and provide new clues for optimizing the methods for its cultivation application.

Usp22 controls multiple signaling pathways that are essential for vasculature formation in the mouse placenta

Project description:Usp22, a component of the SAGA complex, is over expressed in highly aggressive cancers, but the normal functions of this deubiquitinase are not well defined. We determined that loss of Usp22 in mice results in embryonic lethality due to defects in extra-embryonic placental tissues and failure to establish proper vascular interactions with the maternal circulatory system. These phenotypes arise from abnormal gene expression patterns that reflect defective kinase signaling, including TGFβ and several receptor tyrosine kinase (RTK) pathways. Usp22 deletion in endothelial cells and pericytes induced from embryonic stem cells also hinders these signaling cascades with detrimental effects on cell survival and differentiation as well as ability to form vessels. Our findings provide new insights to Usp22 functions during development that may offer clues to its role in disease states.
