Proteome analysis of the spontaneous oscillatory behavior in 1,3-propanediol production from glycerol by Clostridium butyricum

ABSTRACT: To investigate the underlying mechanisms responsible for the observed oscillatory behavior, this study employed TMT technology to perform quantitative proteomic analysis. Samples were collected at five time points during the first oscillation cycle .The functional annotation of proteins was identified according to KEGG pathway analysis. In total, 2059 proteins were identified, of which 1412 proteins were quantitated.

Project description:In this work we present proteomic analysis of Bacillus subtilis 168 trp+ (BSB1) during germination and spore outgrowth. Samples were collected from 14 different time points (from 0 to130 minutes) after inoculation of spores into germination medium. In summary, 2,195 proteins were identified and clustered according to their expression kinetics. Results revealed five specific clusters that were functionally analyzed.

Project description:The aim of this study was to perform comparative gene expression analysis of AIP mutation-positive, AIP mutation-negative familial and sporadic somatotroph tumours to discover the genes/pathways responsible for the aggressive phenotype. Gene expression analysis was performed on 25 pituitary samples (five normal pituitary, six AIPpos GH, three AIPneg GH, four sporadic GH, two AIPneg familial NFPA and five sporadic NFPA adenomas) using the Affymetrix human Gene Chip HG-U133 Plus 2.0 array.

Genotype mixing affects gene expression associated with hygienic behavior

Project description:Analysis used total RNA samples of individual brains of low hygienic strain worker bees that were derived from experimental colonies combined of low hygienic genotypes (L pure colonies) resp. low and high hygienic genotypes (L/H mixed colonies). Four-condition experiment: individuals that did perform hygienic behavior (uncapping) vs individuals that did not perform hygienic behavior (non-hygienic); individuals from L pure colonies vs individuals from L/H mixed colonies. A total of 76 biological replicates was analyzed, subdivided into 24 individuals that did perform hygienic behavior in mixed colonies; 24 individuals that did perform hygienic behavior in pure colonies; 14 individuals that did not perform hygienic behavior in mixed colonies; 14 individuals that did not perform hygienic behavior in pure colonies. Each individual brain RNA sample was divided into two subsamples labeled with cy3 resp. cy5. Loop Design was applied so that each individual brain sample was directly compared to at least two other samples.

The Transcriptome of Photo-Biological Hydrogen Production in the Green Alga Chlamydomonas reinhardtii

Project description:Algal photo-bio hydrogen production, a promising method for producing clean and renewable fuel in the form of hydrogen gas, has been studied extensively over the last few decades. In this study, microarray analyses were used to obtain a global expression profile of mRNA abundance in the green alga Chlamydomonas reinhardtii at five different time points before the onset and during the course of sulphur depleted hydrogen production. The present work confirms previous findings on the impacts of sulphur deprivation but also provides new insights into photosynthesis, sulphur assimilation and carbon metabolism under sulphur starvation towards hydrogen production. For instance, while a general trend towards repression of transcripts encoding photosynthetic genes was observed, the abundance of Lhcbm9 (encoding a major light harvesting polypeptide) and LhcSR1 (encoding a chlorophyll binding protein) was strongly elevated throughout the experiment, suggesting remodeling of the photosystem II light harvesting complex as well as an important function of Lhcbm9 under sulphur starvation. This study presents the first global transcriptional analysis of C. reinhardtii during hydrogen production using five major time points at Peak Oxygen, Mid Oxygen, Zero Oxygen, Mid Hydrogen and Peak Hydrogen. Transcription profiling of Arabidopsis thaliana mutants treated with flg22, Alternaria brassicicola, Methyl jasmonate and Pst DC3000 bacteria

Project description:Gibberellin (GA) promotes plant growth by destabilizing DELLA proteins. DELLA proteins integrate multiple hormonal and environmental stress responses. We investigated the role of GA and DELLA proteins in plant defence. We used microarrays to detail the global programme of gene expression controlled by DELLA proteins and identified distinct classes of differentially regulated genes in response to pathogens, hormones or pathogen elicitors. Experiment Overall Design: Five weeks old short day grown Arabidopsis leaf discs were used to treat with flg22 and samples were collected after 1 hour and 2 hour time points. For Alternaria brassicicola, five weeks old plants were drop inoculated with 4x 5Âµl droplets of Alternaria brassicicola spores (5x105 spores/ml) and samples were collected 3 days post inoculation. Five weeks old plants were infiltrated with Pst DC3000 (2x105cfu/ml) bacteria and samples were collected after 12 hours post infiltration. For methyl jasmonate treatments, five weeks old plants were sprayed with 10ÂµM Methyl Jasmonate solution, covered with plastic bags and samples were collected after one hour.

Tissue compartmentalization and distinct stem cell behavior underlie epidermal homeostasis

Project description:Long-term tissue homeostasis is governed by balanced contribution from adult stem cells. We recently identified Lrig1 as a marker of stem cells in mouse epidermis. Here, we show that Lrig1 expressing cells are molecularly and functionally distinct from previously characterized epidermal stem cell populations. During steady state homeostasis, Lrig1 expressing cells are responsible for the maintenance of the uppermost compartment of the pilosebaceous unit known as the infundibulum. Lineage tracing demonstrates that the epidermis is divided into discrete compartments during homeostasis, each maintained by its own resident stem cells. Compartment boundaries are rapidly broken when stem cell progeny are recruited to sites of injury, where they subsequently change behavior according to the environment. Oncogene activation in Lrig1 expressing cells alters proliferation but auxiliary stimuli are required for rapid tumor growth. Our data demonstrate that stem cell niches are compartmentalized according to altering requirements for tissue replenishment.
