Palmitoylation licenses RIPK1 cytotoxicity in the TNF pathway

ABSTRACT: We identified S-palmitoylation as a licensing modification for RIPK1 kinase activity in the TNF pathway. We found that DHHC5 palmitoylates RIPK1 at Cys257 residue, a process dependent on K63-linked ubiquitination of RIPK1, in response to TNF stimulation.
