Exosomal protein identification from untreated mice and macrophage-depleted mice

ABSTRACT: Exosomes are important mediators for cell-cell communication. In previous study, the clearance of exosomes were markedly delayed in systemic circulation of macrophage-depleted mice. To reveal the possible molecular regulation involved in exosome clearance, we performed exosomal proteomic analysis on MP-Exo from macrophage depleted mice and control ones.

Endothelial cells require miR-214 to secrete exosomes that suppress senescence and induce angiogenesis in human and mouse endothelial cells

Project description:Signalling between endothelial cells, endothelial progenitor cells and stromal cells is crucial for the establishment and maintenance of vascular integrity and involves exosomes, among other signalling pathways. Exosomes are important mediators of intercellular communication in immune signalling, tumour survival, stress responses and angiogenesis. The ability of exosomes to incorporate and transfer mRNAs encoding for ‘acquired’ proteins or miRNAs repressing ‘resident’ mRNA translation suggests that they can influence the physiological behaviour of recipient cells. We here demonstrate that miR-214, a miRNA that controls endothelial cell function and angiogenesis, plays a dominant role in exosome-mediated signalling between endothelial cells. Endothelial cell-derived exosomes stimulated migration and angiogenesis in recipient cells, whereas exosomes from miR-214 depleted endothelial cells failed to stimulate these processes. Exosomes containing miR-214 repressed the expression of Ataxia Telangiectasia Mutated in recipient cells, thereby preventing senescence and allowing blood vessel formation. Concordantly, specific reduction of miR-214 content in exosome-producing endothelial cells abolishes the angiogenesis the angiogenesis stimulatory function of the resulting exosomes. Collectively our data indicate that endothelial cells release miR-214 containing exosomes to stimulate angiogenesis through silencing of Ataxia Telangiectasia Mutated in neighbouring target cells. Gene expression analysis of HMEC endothelial cells exposed to supernatant containing either HMEC derived exosomes (miR-214 high), HMEC derived exosomes depleted of miR-214 (miR-214 low) or containing no exosomes (no exosomes). Each sample was analysed in duplo.

Single-cell RNA sequencing reveals the transcriptional landscape and heterogeneity of macrophages in intestinal tissue of WT and CD163+ macrophage-deficient mice.

Project description:CD163 macrophages, which markedly accumulated in inflammation and tumor tissues possessed markedly immunosuppressive functions on immune cells, especially effective cells such as CD8 cells. However, the mechanism of CD163 macrophages in regulating immune responses remains to be illusive. Using single cell-sequence techniques, we identified CD163 macrophage subset in the colon tissues. Meanwhile, we also characterized CD163 macrophage associated immune cell populations and subpopulations in mouse colon tissues. We found that CD163+ macrophage subset in the colon was related to gut microbiota. Deletion of CD163+ macrophage subset could markedly increase the proportions of the immune cell subsets such as CD8 and NK cells. Mechanistically, gut microbiota mediated CD163+ macrophage subpopulations can down-regulate CD8 and NK cells through inhibitory molecules CD94/NKG2a (Kird1/Kirc1 in mice) and their ligand HLA-E /QA-1 (Human/mice) on the surface of CD163+ macrophages.

Pancreatic cancer exosomes induce pre-metastatic niche formation in the liver

Project description:Pancreatic cancers (PCs) are highly metastatic with poor prognosis, mainly due to delayed detection. We hypothesized that intercellular communication is critical for metastatic progression. Here, we show that PC-derived exosomes induce liver pre-metastatic niche formation in naïve mice and consequently increase liver metastatic burden. Uptake of PC-derived exosomes by Kupffer cells caused transforming growth factor β secretion and upregulation of fibronectin production by hepatic stellate cells. This fibrotic microenvironment enhanced recruitment of bone marrow-derived macrophages. We found that macrophage migration inhibitory factor (MIF) was highly expressed in PC-derived exosomes, and its blockade prevented liver pre-metastatic niche formation and metastasis. Compared to patients whose pancreatic tumors did not progress, MIF was markedly higher in exosomes from stage I PC patients who later developed liver metastasis. These findings suggest that exosomal MIF primes the liver for metastasis and may be a prognostic marker for the development of PC liver metastasis.

Astragaloside IV attenuates macrophage-derived exosomes induced peritoneal fibrosis after inflammatory injury through the miR-204-5p/Foxc1 pathway

Project description:Peritoneal dialysis (PD) is a successful renal replacement therapy for end-stage renal disease that effectively improves the quality of life. Long-term PD causes epithelial mesenchymal transformation (MMT) of peritoneal mesothelial cells, leading to peritoneal fibrosis which reduces the efficiency of PD. Macrophages are considered players in the onset and perpetuation of peritoneal injury. Yet, the mechanisms employed by macrophage-mesothelial cells communication to regulate peritoneal fibrosis are not fully elucidated resulting in lack of disease-modified drugs. This study analyzes the role of macrophage-mesothelial cell communication by intraperitoneal injection of macrophage derived exosomes in PD model rats. These results show that macrophages secrete exosomal miR-204-5p that directly targets Foxc1, leading to the activation of MMT in mesothelial cells. The data also shows that intraperitoneal injection of dissolved AS-IV can improve MMT by altering macrophage derived exosomal miRNAs. This study indicates that intercellular crosstalk between peritoneal macrophages and mesothelial cells is mediated by macrophage derived miR-204-5p-containing exosomes that control the MMT progression, providing AS-IV for prevention and treatment of PD induced peritoneal fibrosis. Our results demonstrate, for the first time, a novel role of the AS-IV on miR-204-5p/Foxc1/β-catenin axis in improving peritoneal fibrosis in vivo and vitro.

microRNA expression profile of exosomes from bone marrow derived macrophage

Project description:MicroRNA has the potential for cross-regulation and functional integration of discrete biological processes during complex physiological events. In this study, we found that the highly expressed microRNAs in exosomes from bone marrow derived macrophage appear to control fibrotic healing response in the tendon. Notably, fibrotic microRNA-21 in mice distributed in the early stage of healing after tendon injury, having the similar expression with mammals. Therefore, we hypothesized the bone marrow derived macrophage secreted miRNAs-containing exosomes play important functions in peritendinous adhesion after tendon injury.

Project description:The macrophages in tomor microenvironment in were alike M2 macrophages which contribute to tumor progression and migration. Since macrophages can secret lots of exosomes and M2 macrophage can induce the imbalance of Treg/Th17 ratio in EOC tumor environment, we want to investage the different expression of miRNA between the exosomes secreted by monocyte and M2 macrophage. Thus to see if the microRNA in exosomes secreted from M2 macrophage paly a big role in the T cell imbalance.

Detection of miRNAs in exosomes released by mouse immature and mature dendritic cells

Project description:Dendritic cells (DCs) are the most potent antigen (Ag)-presenting cells. Whereas immature DCs down-regulate T cell responses to induce/maintain immunological tolerance, mature DCs promote immunity. To amplify their functions, DCs communicate with neighboring DCs through soluble mediators, cell-to-cell contact and vesicle exchange. Transfer of nanovesicles (<100nm) derived from the endocytic pathway (termed exosomes) represents a novel mechanism of DC-to-DC communication. The facts that exosomes contain exosome-shuttle microRNAs (miRNAs), and DC functions can be regulated by exogenous miRNAs, suggest that DC-to-DC interactions could be mediated through exosome-shuttle miRNAs, an hypothesis that remains to be tested. Importantly, the mechanism of transfer of exosome-shuttle miRNAs from the exosome lumen to the cytosol of target cells is unknown. Here, we demonstrate that DCs release exosomes with different miRNAs depending on the maturation of the DCs. By visualizing spontaneous transfer of exosomes between DCs, we demonstrate that exosomes fused with the target DCs, the latter followed by release of the exosome content into the DC cytosol. Importantly, exosome-shuttle miRNAs are functional, as they repress target mRNAs of acceptor DCs. Our findings unveil a mechanism of transfer of exosome-shuttle miRNAs between DCs and its role as a means of communication and post-transcriptional regulation between DCs. The study has analyzed the microRNA content of 4 samples of immature exosomes, 4 samples of matures exosomes, 2 samples of immature bone-marrow-derived DCs, and 2 samples of mature bone marrow-derived DCs.
