Protein coverage improvement for mouse lung tissue by using cysteine modification method

ABSTRACT: protein coverage is improved for both total protein and membrane protein by using powerful cysteine modification (S-aminoethylation) method. We successfully identify about 22% extra total protein and 19% extra membrane protein compared with conventional IAM alkylation method with ultrahigh sensitive orbitrap instrument.

Project description:Endometrial remodeling is required for implantation in all mammalian species. To identify the factors that promote cell proliferation at the implantation site were explored in bovine endometrium and extra-embryonic membrane during the peri-implantation period. Microarray analysis showed that the expression levels of various trophoblast specific genes were higher in the extra-embryonic membrane at gravid horn than in the extra-embryonic membrane at non-gravid horn namely, CSH1, PRPs, PAGs, AIF, Muc-1, etc. Furthermore, EGFR, INHBA and INHBB expression were higher in endometrium of gravid horn. Gene expression profiles were analyzed in the gravid and non-gravid extra-embryonic membrane and endometrium at around Day 30 of gestation

Mir 22-3p enhances recombinant protein production from HEK 293 cell by affecting the expression of HIPK1 and FRAT2

Project description:Improving protein production in human embryonic kidney cells is important structural studies and antibody production. The use of small non-protein coding RNA, such as microRNA, has been an effective method for increasing protein expression. Our high-throughput human microRNA screen in HEK 293 cells previously identified miRNA 22-3p as a promising candidate for increasing the expression of luciferase, couple of membrane proteins and a secreted fusion protein. To explore the mechanisms of this increase in protein expression and to understand the intracellular events, we conducted a gene expression analysis of luciferase- expressing HEK 293 cells transfected with a mir-22-3p mimic and compared with cells transfected with a negative control. Following microarray analysis, down-regulated genes were identified and were cross-referenced with the predicted targets of mir-22-3p and with results from our previous high-throughput siRNA screen. By performing common seed analysis on the possible targets, the list was narrowed to two genes, HIPK1 and FRAT2. These two genes were validated as being involved in improving luciferase production using siRNA and qRT PCR.

Suspended hydrogel culture as a method to scale up intestinal organoids

Project description:Primary tissue-derived epithelial organoids are a physiologically relevant in vitro intestinal model that have been implemented for both basic research and drug development applications. The existing method of culturing intestinal organoids in surface-attached native extracellular matrix (ECM) hydrogel domes is not readily amenable to large-scale culture and contributes to culture heterogeneity. We have developed a method of culturing intestinal organoids within suspended basement membrane extract (BME) hydrogels of various geometries, which streamlines the protocol, increases the scalability, enables kinetic sampling, and improves culture uniformity without specialized equipment or additional expertise. We demonstrate the compatibility of this method with multiple culture formats, and provide examples of suspended BME hydrogel organoids in downstream applications: implementation in a medium-throughput drug screen and generation of Transwell monolayers for barrier evaluation. The suspended BME hydrogel culture method will allow intestinal organoids, and potentially other organoid types, to be used more widely and at higher throughputs than previously possible.
