TMT-10plex analysis of endogenous proteins expressed in HEK-293 cells as a recombinant transporter assay system

ABSTRACT: Tandem-mass-tags (TMT)-10plex proteomics characterized endogenous proteins expressed in HEK-293 cells transiently transfected with control vector or transporter cDNA. Proteomic profiles between mock (n=3), naïve (n=3), and HEK-293 cells transfected with MATE1 cDNA (1, 10, 30, 100 ng) were compared.

OAT10 (SLC22A13) is a transporter highly expressed in renal tubules and transporting organic anions including nicotinate, β-hydroxybutyrate, p-aminohippurate, and orotate. In transport assays using Xenopus oocytes and HEK293 cells, we found that apparent substrate selectivity of OAT10 was different between the expression systems, particularly less pronounced uptake of β-hydroxybutyrate in HEK293 cells. Because functional coupling between transporters may interfere with functional properties of t  ...[more]
