Cerebral organoid proteomics reveals signatures of dysregulated cortical development associated with human trisomy 21

ABSTRACT: This experiment is a label-free multidimensional LC/MS/MS analysis of drug treated cerebral organoids (COs), using Orbitrap Fusion mass spectrometer instrumentation. We generated COs from induced pluripotent stem cells (iPSC) from an individual with DS (female C2-4 line, referred to as T21 hereafter) and an unrelated euploid individual (female H50-1 line, referred to as D21 hereafter). The T21 organoids were treated with the drug harmine. Proteomics was used to identify proteins with differential expression in the harmine treatment and T21 vs euploid comparison.
