Rituximab Treatment Timecourse on Primary B cells.

ABSTRACT: The data describe the treatment of a human Primary B cells. The cells were treated with Rituximab and the full untargeted proteomics and transcriptomics were profiled over a total of 6 timepoints (0,1,2,4,7,15) hours. A validation experiment was repeated as a separate experiment using a subset of the time points.

Samples were collected at 15 and 30 min after Ca2+ addition. For the publication, the experiment was repeated twice, and corresponding timepoints were averaged between the experiments. The first two arrays and last two arrays in this set correspond to the separate experiments.

Variation in gene expression patterns in follicular lymphoma and the response to rituximab

Project description:This SuperSeries is composed of the following subset Series: GSE3646: Follicular lymphoma and normal lymphoid tissue comparisons GSE3647: Follicular lymphoma lymph node Abstract: Analysis of the patterns of gene expression in follicular lymphomas from 24 patients suggested that two groups of tumors might be distinguished. All patients, whose biopsies were obtained before any treatment, were treated with rituximab, a monoclonal antibody directed against the B cell antigen, CD20. Gene expression patterns in the tumors that subsequently failed to respond to rituximab appeared more similar to those of normal lymphoid tissues than to gene expression patterns of tumors from rituximab responders. These findings suggest the possibility that the response of follicular lymphoma to rituximab treatment may be predicted from the gene expression pattern of tumors. Refer to individual Series

Stable gene expression of serial skin biopsies defines patient subsets in diffuse cutaneous systemic sclerosis

Project description:Systemic sclerosis (SSc) shows complex clinical manifestations including progressive skin and internal organ fibrosis. SSc can be divided into 'intrinsic subsets' by gene expression suggesting patient-specific heterogeneity in pathogenesis or temporal evolution of disease. Here we validate these subsets using an independent patient population, and test whether the genes vary over time with patients changing subsets as disease progresses, or if the genes are a stable feature of the patients within each subset. Skin biopsies were analyzed from 13 dSSc patients enrolled in an open label study of rituximab, 9 dSSc patients not treated with rituximab, and 9 healthy controls. These data recapitulate the patient 'intrinsic subsets' described previously with gene expression associated with cell proliferation, inflammatory processes, and a normal-like group. Serial skin biopsies showed consistent and non-progressing gene expression. We were unable to detect significant differences in gene expression before and after rituximab treatment, consistent with an apparent lack of clinical response. Serial biopsies from each patient stayed within the same gene expression subset regardless of treatment regimen or the time point at which they were taken. This demonstrates the intrinsic subsets are an inherent, reproducible and stable feature of SSc that is independent of disease duration. Skin biopsies were analyzed from 13 dSSc patients enrolled in an open label study of rituximab, 9 dSSc patients not treated with rituximab, and 9 healthy controls.
