ABSTRACT: Deep seas marine fungus SST2-VLC fractions. Isolated from soil sediment from South Shetland Trench, Antarctica (-58.90865, -61.122976, ~5100 m). Large scale fermentation on rice medium followed by subsequent methanol extraction. Finally, vacuum liquid chromatography was completed using an increasing gradient from hexane-dichloromethane-methanol.

However, how cells respond to methanol and remodel cellular metabolic network on methanol were not clear. Responses to different sulfur sources reveal the complicated and interlinked sulfur metabolism pathways of a deep-sea sulfate reducing bacterium

Project description:Sulfur metabolism in the deep-sea cold seep has been mentioned to have an important contribution to the biogeochemical cycle of sulfur in previous studies. And sulfate reducing bacteria have also been considered to be a dominant microbial population in the deep-sea cold seep and play a crucial role in this process. However, most of sulfate reducing bacteria from cold seep still cannot be purely cultured under laboratory conditions, therefore the actual sulfur metabolism pathways in sulfate reducing bacteria from the deep-sea cold seep have remained unclear. Here, we isolate and pure culture a typical sulfate reducing bacterium Desulfovibrio marinus CS1 from the sediment sample of the deep-sea cold seep in the South China Sea, which provides a probability to understand the sulfur metabolism in the cold seep.

Project description:Purpose: To explore the differentially expressed genes and pathways in the thermotolerant mutant compare to wildtype. To figure out the contribution of these genes and pathways to high protein expression ability in the mutant. Method: the samples were collected from the 60h fermentation culture. The fermentation medium was BMGY/BMMY with 1%(v/v) methanol as inducer. the cultivation condition was set under 28℃ and 37℃. Results: after data analysis, we found genes related to oxidative stress response were important.

Transcriptomic analysis of Pichia pastoris (Komagataella phaffii) GS115 during heterologous protein production using a high-cell-density fed-batch cultivation strategy

Project description:We investigated gene expression during the bench-scale batch fermentation phase, glycerol feed phase, glycerol-methanol mixture feed (GM) phase, and at different time points following methanol induction using RNA-Seq. We report that the addition of the GM phase may help to alleviate the adverse effects of methanol addition (alone) on P. pastoris cells. Secondly, enhanced upregulation of the mitogen-activated protein kinase (MAPK) signaling pathway was observed in P. pastoris following methanol induction. The MAPK signaling pathway may be related to P. pastoris cell growth, and may regulate the AOX1 promoter via regulatory factors activated by methanol-mediated stimulation. Thirdly, the unfolded protein response (UPR) and ER-associated degradation (ERAD) pathways were not significantly upregulated during the methanol induction period. These results imply that the presence of unfolded or misfolded phytase protein did not represent a serious problem in our study. Finally, the upregulation of the autophagy pathway during the methanol induction phase may be related to the degradation of damaged peroxisomes but not to the production of phytase. This work describes the metabolic characteristics of P. pastoris during heterologous protein production under high-cell-density fed-batch cultivation. We believe that the results of this study will aid further in-depth studies of P. pastoris protein expression, regulation, and secretory mechanisms.
