ABSTRACT: we analyzed the metabolome content by untargeted HPLC/HRMS (see supplementary information). Isolates were first grown in synthetic CF medium 2 (SCFM2), a culture medium designed to mimick the nutritional conditions in CF pulmonary mucus (Turner et al. 2014). Intracellular metabolic extracts of mid-log cultures were harvested by fast-filtration and mechanical lysis, and the contents were normalized to a constant colony-forming units over optical density at 595 nm ratio (CFU/DO), as described by Aros-Calt et al. (Aros-Calt et al. 2019). Metabolomics analysis were performed using two complementary HPLC/HRMS methods to optimize chemical coverage, and data were processed and normalized following the most up-to-date methods
