Project description:Tuberculosis (TB) remains a major public health problem and we lack a comprehensive understanding of how Mycobacterium tuberculosis (M. tb) infection impacts host immune responses. We compared, at two timepoints, the induced immune response to TB antigen, BCG and IL-1β stimulation between latently M. tb infected individuals (LTBI) and active TB patients. The immune response was assessed using the TruCulture system with a Null stimulation. samples were tested by Ultrahigh Performance Liquid Chromatography-Tandem Mass Spectroscopy (UPLC MS/MS) for a total of 696 metabolites.

Transcriptional and proteomic insights into phytotoxic activity of interspecific potato hybrids with low glycoalkaloid contents.

Project description:We examined phytotoxic potential of diploid potato hybrids from 15-1 population. Individuals were bulked in bulks C and D. Both bulks characterized low total glycoalkaloids content and differ phytotoxic potential. Water extract prepared from leaves of individuals of bulk C inhibited growth of test plant - mustard in 40%, while in bulk D no inhibition was observed. To evaluate cause of expression of phytotoxic potential in bulk C, we examined glycoalklaoids composition in leaf water extract (mass spectrometry), compared gene (deep RNA sequencing - BGISEQ) and protein expression (nano-LC-MS-MS/MS) profiles between the bulks. Bulks differed in glycoalkaloids composition, bulk C contained alpha-solasonine which wasn't detected in bulk D. We concluded, alpha-solasonine may act together with alpha-solamargine and increases phytotoxic potential of leave extract of bulk C. Comparison of gene expression profiles indicating on contribution of genes, not directly involved in glycoalkaloids biosynthesis to express phytotoxic potential of C bulk. We have found 3 proteins characteristic for C bulk and 2 proteins characteristic for D bulk. We found one protein threonine dehydratase biosynthetic, which is more abundant in D bulk and his abundance corresponds with gene transcripts found in RNA-seq experiment.

Effets of plant compounds on estrogen receptor positive breast cancer cell line MCF-7

Project description:This study was designed to determine the effects of plant derived compounds on the transcriptome of estrogen receptor positive breast cancer. Phytoestrogens are compounds produced by plants that possess weak estrogenic activity. They can be found in a wide variety of plants, including fruits and vegetables, but are most abundant in leguminous plants. Legumes are consumed in almost every diet throughout the world, and in addition to the seeds, many other parts of the plant are also edible, including the pods of some varieties. Soybeans have been demonstrated to contain high concentrations of the isoflavones daidzein and genistein, which are re-sponsible for many of the health benefits of soy. Besides isoflavonoids, flavonoids also exert estrogenic activity, but usually at a much lower level of activity compared to that of isoflavonoids. Also, some flavonoids, including kaempferol and quercetin, canexhibit antiestrogenic activity, with several legumes being a source of these flavo-noids. Coumestrol, a coumestan with high estrogenic activity in cell and animal as-says, is also present in several legume seeds and sprouts. These underutilized plant compounds could hold previously unknown potential for antioxidant activity, anti-inflammation activity, cholesterol lowering ability, and even anticancer activity. The goal of the present study was to identify the effects of plant derived compounds on estrogen receptor positive cell line to determine if the compound possess estrogenic or anti-estrogenic activity, thereby highlighting potential implications on human health through diet.

Nitrophenol (m-nitrophenol) treatment with 5 mM for 2 h

While these industrial chemicals provide numerous benefits, there is no doubt that some have potential to damage the environment and health. Toxicity must be evaluated and use must be carefully controlled and monitored in order to minimize potential damage. DNA microarray technology has become an important new technique in toxicology. We are using the yeast Saccharomyces cerevisiae as a model organism for toxicological study because it is a simple, fast-growing eukaryote that has been thoroughly characterized. In order to evaluate toxicity by newly synthesized or mixture chemicals, toxicity-induced gene expression alteration profiles by known chemicals should be collected. In our study, cells need to be exposed with same experimental cellular condition, semi lethal (IC50), respectively. In the case of m-nitrophenol (CAS; 554-84-7), the exposure dose was decided as 5 mM by growth curve with continuously diluted exposure. m-Nitrophenol is one of the nitrogen compounds. Human cerebrospinal fluid autoantibody lipid microarray profiling (Fig. 2C)

Project description:Lipids comprise 70% of the myelin sheath, and autoantibodies against lipids may contribute to the demyelination that characterizes multiple sclerosis (MS). We used lipid antigen microarrays and lipid mass spectrometry to identify bona fide lipid targets of the autoimmune response in MS brain and an animal model of MS to explore the role of the identified lipids in autoimmune demyelination. We found that autoantibodies in MS target a phosphate group in phosphatidylserine and oxidized phosphatidylcholine derivatives. Administration of these lipids ameliorated experimental autoimmune encephalomyelitis by suppressing activation and inducing apoptosis of autoreactive T cells, effects mediated by the lipids' saturated fatty-acid side chains. Thus, phospholipids represent a natural anti-inflammatory class of compounds that have potential as novel therapeutics for MS.
