ABSTRACT: Control bone extractions completed using an SP3 based extraction and used to confirm the sharred masses in collagen peptide mass fingerprints. Project description:Genotypes used are Arabidopsis thaliana (L.) Col-0 wild-type, atg5.3 mutant (SALK_020601), sid2.2 (Dewdney et al., 2000) and the atg5.3/sid2.2 double mutant that has been previously characterized by Yoshimoto et al. Plant Cell (2009). Plants were cultivated under control conditions (ideal nutrition N+S+; 10mM NO3- and 0.266mM SO42-), low nitrate (N-; 2 mM NO3- and 0.266mM SO42-), or low sulfate (S-; 10mM NO3- and 0.016mM SO42-) conditions, under in short days (160 micromol.m-2.s-1; 8 h light, 16 h dark) in growth chamber as described by Havé et al. 2019 New Phytologist. Rosettes were harvested at vegetative stage 60 days after sowing, 2-3 hours after the beginning of the photoperiod. Two consecutive cultures (C1 and C2) were performed at several month interval. For each culture, three plant repeats (R1-R3) were collected, each repeat was a mix of >6 rosettes. Details can be found in Havé et al. (2019) New Phytologist (doi: 10.1111/nph.15913). Dual knockdown of Esrrb and Sox2 in mouse embryonic stem cells

Project description:Esrrb, Sox2 or Esrrb and Sox2 were knocked down in mouse ES cells using shRNA plasmid pSUPER.puro containing shRNAs from the publications Feng et al., 2009 and Rodda et al., 2005. Knockdown was transfected into mouse ES cells using lipofectamine and then cells were selected for knockdown using puromycin. RNA was harvested after 2 days of knockdown.

Role of the mRNA-binding protein HuR in the global transcriptomic status of cells stimulated with electrophiles and oxidants

Project description:HuR has been implicated as a stress-responsive protein: cytoplasmic translocation of HuR is promoted by several small-molecule stressors, e.g., H2O2, arsenite, and the cyclopentenone prostaglandin A2 (PGA2), a Michael acceptor RES (Wang et al., 2000; Yang et al., 2004). Treatment with PGA2 also increases the affinity of HuR for p21 mRNA (Wang et al., 2000). Intrigued by these reports, we launched HuR-dependent gene-expression profiling studies in the presence and absence of small-molecule stress signals. HNE—a native RES with a reactive core chemically similar to that of PGA2—was selected as a representative bioactive RES (Jacobs and Marnett, 2009; Schopfer et al., 2011) and H2O2 was selected as a representative ROS. shHuR and shControl HEK293T cells were first generated and then treated with HNE or H2O2 under the conditions where cell viability is largely unaffected. We subsequently sequenced their RNA post ribosomal RNA (rRNA) depletion (see details below). Significant differential expression was evaluated with CuffDiff (Trapnell et al., 2013).

Comparing the effects of live versus recorded relaxation on mood, coping, and quality of life on patients with cancer

Project description:Study hypothesis: 1. In patients attending the Oncology Health Centres for the first time, what is the relative effectiveness of live relaxation "LIVEREL" and recorded relaxation "RECREL" (i.e. "treatment as usual" including audio recorded relaxation as part of support within the Oncology Health Centres) on mood, coping and quality of life? 2. What is the relative cost-effectiveness of LIVEREL and RECREL? 3. What are the immediate effects of live versus recorded relaxation instructions on blood pressure and pulse rate? If patients randomised to LIVEREL have significantly higher relaxation scores on the Mood Rating Scale than those randomised to RECREL, we would conclude that the beneficial effects of LIVEREL was due to the extra social contact and/or the effects of live feedback on performance. Primary outcome(s): Relaxation scale score on the Mood Rating Scale (MRS), (Anderson et al., 2000; Johnson et al., 1996; Walker et al., 1997; Walker et al., 1999; Wesnes et al., 1997b), measured at 8 weeks

Comprehensive Microarray Transcriptome Profiling of CD34-Enriched Mouse Keratinocyte Stem Cells

Project description:Recent developments in animal models (Morris et al., 2004; Tumbar et al., 2004) as well as the discovery of cell surface markers (Jones and Watt, 1993; Tani et al., 2000; Trempus et al., 2003; Nijhof et al., 2006) have made it possible to isolate living epidermal hair follicle stem cells (HFSCs) from mouse skin, facilitating the study of the biological and molecular features inherent to HFSCs. A complexity of stem and progenitor cell populations within the hair follicle has been revealed. Here, we report comprehensive profiling of mouse CD34-expressing HFSCs using the Agilent mouse oligo microarray platform in order to extend and enrich the existing HFSC databases.
