Twinfilin1 controls lamellipodial protrusive activity and actin turnover during vertebrate gastrulation

ABSTRACT: In this paper, using the Xenopus dorsal gastrula mesoderm, we explored the localization dynamics of several actin-regulatory proteins, defined the in vivo interactome of Cofilin2 (CFL2), and explored the function of Twinfilin1 in convergent extension. Specifically, this APMS dataset is comprised of "prey" protein interactions determined to associate with a CFL2-GFP "bait."

Project description:Explanted tissues from vertebrate embryos reliably develop in culture and have provided essential paradigms for understanding embryogenesis, from early embryological investigations of induction, to the extensive study of Xenopus animal caps, to the current studies of mammalian gastruloids. Cultured explants of the Xenopus dorsal marginal zone (“Keller” explants) serve as a central paradigm for studies of convergent extension cell movements, yet we know little about the global patterns of gene expression in these explants. In an effort to more thoroughly develop this important model system, we provide here a time-resolved bulk transcriptome for developing Keller explants.

Cell adhesions link membrane scale protein dynamics to tissue scale force production during vertebrate head-to-tail axis extension

Project description:In this paper, we use approaches from proteomics, cell biology, and tissue biomechanics to describe how a poorly characterized cell adhesion effector, Armadillo Repeat protein deleted in Velo-Cardio-Facial syndrome (ARVCF) catenin, controls vertebrate head-to-tail axis extension. We define the in vivo interactome of c-Cadherin (Cdh3), measuring its interaction with Arvcf. Specifically, this APMS dataset is comprised of "prey" protein interactions determined to associate with a Cdh3-GFP "bait."

Identification of specific xWNT5A and xWNT11specific target genes in dorsal marginal zone explants of Xenopus laevis

Project description:The non-canonical signaling branches of the WNT signaling network realize different movements during Xenopus laevis gastrulation movements. The WNT5A branch is responsible for the constriction whereas the WNT11 branch triggers the elongation of the dorsal mesoderm. Therefore we used dorsal marginal zone explants which undergo convergent extension movements as readout system to identify differences in gene expression pattern between the WNT5A and WNT11 signaling branch via microarray analysis.We identified pbk as specific WNT5A target gene and rab11fip5 as specific WNT11 target gene. Therefore the non-canonical branches of the WNT signaling network do not only regulate cell behavior but also the expression of different target genes.

dact1/2 modifies noncanonical Wnt signaling and calpain 8 expression to regulate convergent extension and craniofacial development

Project description:Wnt signaling plays a fundamental role in the initial patterning and development of the embryo, including in the regulation of convergent extension during gastrulation and the establishment of the dorsal axis. Further, Wnt signaling is a crucial regulator of craniofacial morphogenesis. The relationship between early embryo patterning and craniofacial outcomes warrants further study. The adapter proteins Dact1 and Dact2 modulate the Wnt signaling pathway through binding to Disheveled, however, the distinct roles of Dact1 and Dact2 during embryogenesis remain to be fully elucidated. In this study, we investigated the spatiotemporal gene expression patterns of dact1 and dact2 during zebrafish embryogenesis, revealing both shared and unique domains of expression. We found that both dact1 and dact2 contribute to axis extension, with compound mutants exhibiting a similar convergent extension defect and craniofacial phenotype to the wnt11f2/slb mutant. Utilizing single-cell RNAseq and gpc4-/- zebrafish, a convergent extension mutant with an opposite craniofacial phenotype, we identified dact1/2 specific roles during early development. Using this subtractive approach, we discovered a novel role for dact1/2 in regulating the mRNA expression of the classical calpain, capn8, suggesting a previously unappreciated role of calcium-dependent proteolysis during embryogenesis. Taken together, our findings highlight the distinct and overlapping roles of dact1 and dact2 in embryonic craniofacial development, providing new insights into the multifaceted regulation of Wnt signaling.

Neuroligin-associated microRNA-932 targets actin and regulates memory

Project description:Increasing evidence suggests microRNAs (miRNAs) control levels of mRNA expression during development of the nervous system and during sensory elicited remodelling of the brain. We used an associative olfactory learning paradigm (proboscis extension response) in the honeybee Apis mellifera to detect gene expression changes in the brain. Transcriptome analysis of bees trained to associate an odor with a reward and control bees exposed to air without reward, helped us abstract mRNA-miRNA interactions for empirical testing. Functional studies, feeding cholesterol-conjugated antisense RNA to bees resulted in the inhibition of miR-210 and of miR-932 that is embedded within the neuroligin 2 (Nlg2) gene involved in synapse development. Loss of miR-932 prevents long-term memory formation but not learning. We validated 3M-bM-^@M-^YUTR target site interactions of miR-932 and show miR-932 dysregulates actin, a key cytoskeletal molecule involved in neuronal development and activity-dependent plasticity of the brain. The analysis used Air group (no odor learning) as control sample for comparison to two groups of odor-conditioned bees: linalool and floral mix.

A novel Golgi-associated Vangl2 Translational variant is required for PCP regulation in vertebrates

Project description:First described in Drosophila melanogaster, planar cell polarity (PCP) is a developmental process essential for embryogenesis and development of polarized structures in Metazoans. This signaling pathway involves a set of evolutionarily conserved genes encoding transmembrane (Vangl, Frizzled, Celsr) and cytoplasmic (Prickle, Dishevelled) molecules. Vangl2 is of major importance in embryonic development as illustrated by its pivotal role during neural tube closure in human, mouse, Xenopus and zebrafish embryos. The regulated and poorly understood traffic of Vangl2 to the plasma membrane is a key event for its function in development. Here we identify a novel N-terminally extended isoform of Vangl2, termed Vangl2-Long that arises from the use of non-AUG start codon upstream of the coding region of canonical Vangl2. Vangl2-Long contains an evolutionarily conserved N-terminal 48 amino acid sequence bearing a signal for subcellular localization in the Golgi apparatus. Moreover, we provide data in Xenopus showing that Vangl2-Long is important for correct convergent extension movements and neural tube closure. These data describe a further level of complexity in Vangl2 expression, trafficking and function.

Identifying the targets of the pair-rule transcription factors Even-skipped and Runt in the late Drosophila blastoderm

Project description:It has long been appreciated that striped pair-rule transcription factor expression is necessary for convergent extension in the early Drosophila embryo, although the mechanisms that link these transcriptional regulators to planar polarity in this tissue have long been elusive. The goal of this study was to determine the transcriptional tragets of the pair-rule transcription factors Eve and Runt in Drosophila blastoderm embryos. We compared the transcriptional profiles of late blastoderm embryos injected with either water or dsRNAs against both eve and runt to identify differentially expressed genes that may directly contribute to the establishment of planar polarity during Drosophila convergent extension. Comparing the mRNA profiles from late blastoderm Drosophila embryos injected with either water (Water) or eve+runt dsRNAs (Eve), in triplicate, using Illumina HiSeq.

We characterized the early, intermediate, and late stages of Xenopus tropicalis tail regeneration using the Xenopus tropicalis Affymetrix genome array in biological replicate.
