Phosphoproteome analysis in Arabidopsis thaliana when exposed to immunogenic molecular patterns

Project description:In plants, recognition of immunogenic molecular patterns, such as bacterial flagellin (flg22 epitope), leads to an enhanced state of immunity, designated pattern-triggered immunity (PTI). Following cognate ligand perception, pattern recognition receptors initiate sequential phosphorylation events to activate defense responses against invading pathogens. To gain further insight into PTI signaling, we conducted phosphoproteome analyses in Arabidopsis seedlings with immunogenic molecular patterns.

Convergent evolution of plant pattern recognition receptors sensing cysteine-rich patterns from three microbial kingdoms

Project description:Plants deploy pattern recognition receptors to detect microbe- and damage-associated molecular patterns. Arabidopsis thaliana receptor-like protein RLP30 contributes to innate immunity to the necrotrophic fungus Sclerotinia sclerotiorum by recognizing SCLEROTINIA CULTURE FILTRATE ELICITOR 1 (SCFE1). Here we show that the S. sclerotiorum small cysteine-rich protein SCP1 accounts for elicitor activity of SCFE1. RLP30 recognizes SCP1 and its homologs from divergent fungi and oomycetes, as well as an SCP1-unrelated and conserved pattern from bacterial Pseudomonads. Stable expression of RLP30 in Nicotiana tabacum confers enhanced immunity to bacterial, fungal, and oomycete pathogens. Unlike Arabidopsis, which requires intact SCP1 for RLP30-mediated immunity, other Brassicaceae and Solanaceae respond to smaller immunogenic SCP1 epitopes. We conclude that Arabidopsis RLP30 recognizes immunogenic patterns from three microbial kingdoms and that mechanistically different SCP1 perception has evolved in other plant species, likely as a result of convergent evolution.

Accordingly, it becomes increasingly clear that DAMPs play important role in pathogenesis of multiple diseases including inflammatory diseases and cancer. To get a comprihensive view of DAMP-mediated inflammatory responses, we perfomed microarray analysis on murine macrophages treated by dead cell-derived molecules.

Effect of S4 treatment on glioma cells

Project description:S4, a sulfonamide drug, has been confirmed to induce apoptosis and autophagy in cancer cells. Immunogenic cell death is a special cell death type which is closely related to apoptosis and autophagy. We performed RNA-seq to determine the impact of S4 on global gene expression profile in LN229 cells. Our results show that S4 induces immunogenic cell death via the response to endoplasmic reticulum stress.

A pooled treatment-curated breast cancer gene-expression dataset

Project description:Pre-clinical studies reported the immunogenic or immunomodulatory effects of traditional cancer therapies. However, the publicly available well-curated and harmonized breast cancer datasets, such as TCGA, METABRIC, and MetaGxBreast, lack careful curation of treatment regimens. Hence, limited exploration of the impact of therapies on the prognostic/predictive value of breast cancer biomarkers. Herein, we describe a pooled and treatment-curated gene-expression dataset to investigate the impact of treatments on the prognostic/predictive value of biomarkers. We searched the gene expression omnibus database to identify potential human breast cancer gene-expression datasets with anthracycline/taxane treatment. Published datasets with the detailed treatment regimen, clinical endpoint, clinical-pathological, and gene-expression data were extracted and harmonized. The dataset described herein would help researchers explore the interaction between gene-expression biomarkers and immunogenic/immunomodulatory treatments in breast cancer.

Polycomb group protein BMI1 prevents neuroblastoma cells from DNA-damage-induced apoptotic cell death

Project description:BMI1 is a polycomb protein and its overexpression has been correlated with cancer development and aggressiveness. We previously reported that v-myc avian myelocytomatosis viral oncogene neuroblastoma-derived homolog (MYCN)-induced BMI1 positively regulated neuroblastoma (NB) cell proliferation via the transcriptional suppression of tumor suppressors in NB cells. In order to evaluate the potential of BMI1 as a new target for NB therapy, we herein examined the effects of BMI1 reductions on NB cell differentiation and apoptotic NB cell death. BMI1 knockdown (KD) up to 7 days in NB cells significantly induced their differentiation. BMI1 depletion up to 14 days significantly induced apoptotic NB cell death along with the activation of p53, increases in p73, and induction of p53 family downstream molecules. BMI1 depletion in vivo markedly suppressed NB xenograft tumor growth. A pathological analysis using the TUNEL assay indicated the induction of apoptotic cell death. BMI1 reductions activated ATM and increased γ-H2AX in NB cells. Importantly, these DNA damage signals and apoptotic cell death were not canceled by transduction of polycomb group molecules EZH2 and RING1B. Further, EZH2 or RING1B knockdown could not induce apoptotic NB cell death at the same level of BMI1 KD. Together, BMI1 appears to be a promising target of molecular therapy for NB tumors and our report clarified, for the first time, the shared role of PcG proteins in DNA damage response of NB cells.
