Proteomics and ultrastructural analysis of Hermetia illucens (Diptera: Stratiomyidae) larval peritrophic matrix

ABSTRACT: This is a set of three raw data from the larval of Hermetia illucens. The total protein was extracted with protein lysis buffer (8M urea with appropriate protease inhibitor). The trypsin-digested peptides were analyzed by online nano-flow liquid chromatography tandem mass spectrometry performed on an EASY-nLC system (Thermo, USA) connected to a Q Exactive HF-X Quadrupole Orbitrap mass spectrometer (Thermo, USA) through a nanoelectrospray ion source.

Scallop byssal protein identification

Project description:To explore the protein components for scallop byssus, the soluble fractions of scallop byssus was extract. For mass spectrometric analysis, proteins were extracted from byssal adhesive plaques, and the major SDS-PAGE fractions was treated with trypsin and analyzed using an Easy-nLC nanoflow HPLC system connected to an Orbitrap Elite mass spectrometer (Thermo Fisher Scientific, USA). The mass spectrometry raw data were searched against the full set of predicted proteins from the C. farreri genome using Mascot v2.3.0 (Matrix Science, London, UK).

Mass spectrometry to screen potential ORMs-interacting proteins

Project description:To screen potential ORMs-interacting proteins, HEK293T cells were transfected with plasmids containing HA-tagged ORM1, HA-tagged ORM2, HA-tagged ORM3 and HA-Empty vector, respectively. Then, protein lysates were harvested for immunoprecipitation. Mass spectrometry combined with high-performance liquid chromatography (HPLC-MS) was performed by Cloud‐Seq Biotech (Shanghai, China) using Q Exactive Orbitrap Mass Spectrometer (Thermo Fisher Scientific, Waltham, MA, USA). Proteins were digested with trypsin (trypsin: protein = 1:50 (w/w)) at 37°C for 16 hours. After digestion, the protein fragments were separated by high pH reverse-phase HPLC. The peptides were then analyzed by an EASY-nLC 1000 system connected to an Q Exactive mass spectrometer and Orbitrap Fusion mass spectrometer (Thermo Fisher Scientific, Waltham, MA, USA). In raw file, MaxQuant 1.6.0.16 was used to analyze the data, and MaxQuant own algorithm was used to identify peptides and proteins.

Analysis of Erythroleukemia

Project description:Samples were subjected to LC–MS analysis using a dual pressure LTQ-Orbitrap Elite mass spectrometer (Thermo Fisher Scientific) connected to an electrospray ion source (Thermo Fisher Scientific) as recently described (PMID: 23017020). Peptide separation was carried out on an EASY nLC-1000 system (Thermo Fisher Scientific) equipped with a RP-HPLC column (75 μm × 30 cm) packed in-house with C18 resin (ReproSil-Pur C18–AQ, 1.9 μm resin, Dr. Maisch GmbH). A step-wise gradient from 95% solvent A (0.1% formic acid) and 5% solvent B (80% acetonitrile, 0.1% formic acid) to 50% solvent B over 60 min at a flow rate of 0.2 μl/min was used. Data acquisition mode was set to obtain one high resolution MS scan in the FT part of the mass spectrometer at a resolution of 240,000 full width at half-maximum (at m/z 400) followed by 20 MS/MS scans (TOP20) in the linear ion trap of the most intense ions using rapid scan speed. Unassigned and singly charged ions were excluded from analysis. Dynamic exclusion duration was set to 30 seconds.
