Proteomic analysis of Fusarium sp. DS 682 Fungal Mineral Weathering in a Mineral Doped Soil Micromodel


ABSTRACT: Cell lysates of Fusarium sp. DS 682, a saprotrophic fungus, were prepared from fungal biomass grown on PDA agar without M9 and micronutrients. Fungus was exposed to minerals, 100 uL of 50% (w/v) natural kaolinite solution. Three fungal plate collections for each growth condition, with (+ mineral) and without mineral (- mineral), were grown for 14 days on PDA (incubated at 28 C) and harvested after 15 days. Extracted hyphae (500 mg) was prepared using the MPLEx protocol. A nanoACQUITY ultra performance liquid chromatography (LC) with a 2DLC system was used for separation of protein digests. Eluted peptides from the C18 column were analyzed using a Q-Exactive Plus Orbitrap MS for high resolution MS and high-energy collision-induced dissociation tandem MS by electrospray ionization for subsequent quantitative proteomic analysis. Soil fungi facilitate the translocation of inorganic nutrients from soil minerals to other microorganisms and plants.
