VVC-associated strains of Candida albicans provoke more fungal shedding and exfoliation than colonizing strains in epithelial cell infections

Project description:Vulvovaginal candidiasis (VVC) affects nearly ¾ of women during their lifetime and its symptoms seriously reduce quality of life. Although Candida albicans is a common commensal, it is unknown if VVC results from a commensal to pathogenic state switch, if only some strains can cause VVC, and/or if there is displacement of commensal strains with more pathogenic strains. We studied a set of VVC and Colonizing C. albicans strains to identify consistent in vitro phenotypes associated with one group or the other. We find that the strains do not differ in overall genetic profile or behavior in culture media (i.e. MLST profile, rate of growth, filamentation), but they show strikingly different behavior during their interactions with vaginal epithelial cells. Epithelial infections with VVC-derived strains yielded stronger fungal proliferation and shedding of fungi and epithelial cells. RNA-seq analysis of representative epithelial cell infections with selected pathogenic or commensal isolates identified several differentially activated epithelial signaling pathways, including the integrin, ferroptosis and type I interferon pathways; the latter has been implicated in damage protection. The type I interferon pathway is activated more by the Colonizing strains, while type I interferon inhibition selectively increases fungal shedding of only the colonizing fungi. These data suggest that VVC strains may intrinsically have more pathogenic potential via differential elicitation of epithelial responses, including type I interferon pathway. Therefore, it may be possible to evaluate pathogenic potential in vitro to refine VVC diagnosis.

Project description:EXPERIMENT 1: identify commonalities and differences between species at late log phase. EXPERIMENT 2: identify commonalities and differences within a species in function of the media used for the subset of bacteria cultured in more than one media. EXPERIMENT 3: identify commonalities and differences within a species in function of the growth phase for the subset of strains for which more than one time point was taken during the growth phase. Factor 1: growth phase for this slow growers as defined by early, mid and late log phase. Factor 2:rich media used for growth which are either NOS or OMIZ supplemented or not of TPP (thiamine pyrophosphate) a required supplement for some strains. Factor 3: cell pellet and supernatant of culture in which bacetria excrete small molecules. Factor 4. the bacterial species. All those bacteria come from the same genus and have been isolated in function of inflmmatory conditions in human.

Fungal consortium of two Beauveria bassiana strains increases their virulence, growth, and resistance to stress: a metabolomic approach.

Project description:Entomopathogenic fungi have been successfully used to control agricultural pests. They infect insects by coming into direct contact with their cuticle or when feeding on contaminated leaves or fruits. After contact with the insect, the entomopathogenic fungus penetrates its body cavity, where it grows and colonizes it from within, causing its death The use of two or more microorganisms in a microbial consortium has been increasingly applied in the biological control of diseases and pests. Beauveria bassiana is one of the most widely studied fungal species in biological control, yet little is known about its role in fungal consortiums. In a previous study, our group found that a consortium formed by two strains of B. bassiana had significantly greater biocontrol potential against the polyphagous caterpillars Duponchelia fovealis (Lepidoptera: Crambidae) than either strain on its own. Despite recent developments and growing efforts to better understand fungal metabolism and metabolites, much remains unknown. Metabolomics therefore represents an important field for evaluating the metabolites produced or modified by an organism or its relationship with the environment. In the present study, we aim to use untargeted metabolomics with gas and liquid chromatography coupled to mass spectrometers (GC-MS and LC-MS/MS) to evaluate the metabolic alterations caused by the co-cultivation of these strains and to correlate the metabolites produced by this consortium with the increased mortality in D. fovealis observed previosly.

Expression profiling of wild-type and set3D fission yeast strains

Project description:In fission yeast the SET domain protein, Set3p is required for the reliable execution of cytokinesis. To address whether the deletion of the set3 gene might selectively alter expression of cytokinesis genes, expression profiling of wildtype and set3D strains was performed. Wild-type or set3D strains were grown to mid-log phase in YES media and treated with 0.5 mM LatA (or the solvent control, DMSO) for three hours at 30 degrees C. Three biological replicates were performed.

Coordination of fungal biofilm development by extracellular vesicle cargo

Project description:The fungal pathogen Candida albicans can form biofilms that protect it from drugs and the immune system. The biofilm cells release extracellular vesicles (EVs) that promote extracellular matrix formation and resistance to antifungal drugs. Here, we define functions for numerous EV cargo proteins in biofilm matrix assembly and drug resistance, as well as in fungal cell adhesion and dissemination. We use a machine-learning analysis of cargo proteomic data from mutants with EV production defects to identify 63 candidate gene products for which we construct mutant and complemented strains for study. Among these, 17 mutants display reduced biofilm matrix accumulation and antifungal drug resistance. An additional subset of 8 cargo mutants exhibit defects in adhesion and/or dispersion. Representative cargo proteins are shown to function as EV cargo through the ability of exogenous wild-type EVs to complement mutant phenotypic defects. Most functionally assigned cargo proteins have roles in two or more of the biofilm phases. Our results support that EVs provide community coordination throughout biofilm development in C. albicans.
