Temporal Proteomic and Phosphoproteomic Analysis of EV-A71-infected Human Cells

ABSTRACT: We performed time-resolved quantification of the whole proteome and phosphoproteome changes of rhabdomyosarcoma cells (RD) upon EV-A71 infection at 0 hpi, 1 hpi, 2 hpi, 4 hpi, 8 hpi, and 12 hpi based on 6-plex TMT labeling method. This project contains all LC-MS/MS raw files.

Global quantitative proteomic analysis of human glioma cells profiled host protein expression in response to enterovirus type 71 infection

Project description:Enterovirus 71 (EV71) is one of the leading causes of hand, foot and mouth disease with neurological complications in some cases. To study the pathogenesis of EV71 infection, large scale analyses of EV71 infected cells have been performed. However, most of these studies employed rhabdomyosarcoma (RD) cells or used transcriptomic strategy. Here, we performed SILAC-based quantitative proteomic analysis of EV71-infected U251 cells, a human glioma cell line. A total of 3,125 host proteins were quantified, 451 of which were differentially regulated as a result of EV71 infection at 8 hpi or 20 hpi or both.

Project description:We report here the analysis of TRPS1 chromatin binding by ChIP-Seq in embryonal rhabdomyosarcoma RD cells. We use here parental and TRPS1-KO RD cells. RD cells and RD-TRPS1-KO cells were cultured in growth medium and chromatin was prepared. TRPS1 DNA binding in RD cells and RD-TRPS1-KO cells using a selfmade antibody (referenced in Elster et al.) was determined by ChIP-seq.
