Transcriptome sequencing and expression analysis of green sulcus and yellow culm in Phyllostachys violascens cv. Viridisulcata

Project description:In this study, we performed transcriptomics across P. violascens to identify the main pigments contributing to colorful culm and try to reveal a plausible transcriptional regulatory mechanism. The green sulci and yellow culms were collected from the middle of three bamboo’s longest internodes and the chosen bamboos were approximately in the same size which were both 3 meters high, in April 24, 2019. A total of 1,163,327,390 clean reads representing a total of 174.49 Gb nucleotides were generated, with an average GC content of 48.03%. A total 421 differentially expressed genes were identified in the pool.

Continuous Glucose Monitoring for Colorectal Cancer

Project description:Hyperglycemia commonly occurs during surgery due to a reaction to metabolic stress and trauma. It has been shown that improper glycemia control leads to impaired wound healing and a higher risk of other postoperative complications. The primary aim of our project is to assess the feasibility of the use of continuous glucose monitoring in measuring blood glucose levels in patients undergoing colorectal cancer surgery. The secondary aim is to analyze changes in perioperative blood glucose levels to understand the effects of stress and intraoperative interventions on the blood glucose level. The tertiary goal is to assess the predictive value of hyperglycemia for surgical site infection.

Project description:We examined miRNAs expression profiles in rat hearts. The expression of five of 350 scanned miRNAs significantly differed between rat hearts exposed to vehicle and diabetic rat hearts. The expression of 31 miRNAs significantly differed between vehicle and diabetes with fluctuations in glucose levels, and that of 22 miRNAs significantly differed between diabetic hearts and diabetic hearts with glucose fluctuations.

Project description:To analyze the effect of global translation initiation inhibition associated to glucose deprivation on cytoplasmic lncRNAs levels, we performed RNA-Seq using WT, xrn1-delta and upf1-delta cells grown in glucose-containing complete synthetic medium (CSM) and then shifted for 16 min in glycerol- and ethanol-containing medium. In parallel, control cells were maintained for the same time in glucose-containing CSM. For the WT strains, we also included a treatment of 15 min with CHX (100 μg/ml final concentration) or an equal volume of DMSO (Mock).

The secretome and N-glycosylation profiles of the charophycean green alga, Penium margaritaceum, resemble those of embryophytes

Project description:The secretome is defined as the population of proteins that are secreted into the extracellular environment. Many proteins that are secreted by eukaryotes are N-glycosylated; however, there are striking differences in the diversity and conservation of N-glycosylation patterns between taxa. For example, the secretome and N-glycosylation structures differ between land plants and chlorophyte green algae, but it is not clear when this divergence took place during plant evolution. A potentially valuable system to study this issue is provided by the charophycean green algae (CGA), the immediate ancestors of land plants. In this study, we used lectin affinity chromatography (LAC) coupled with mass spectrometry to characterize the secretome, including secreted N-glycoproteins, of Penium margaritaceum, a member of the CGA. The identified secreted proteins and N-glycans were compared to those known from the chlorophyte green alga Chlamydomonas reinhardtii and the model land plant, Arabidopsis thaliana, to establish their evolutionary context. Our approach allowed the identification of cell wall proteins and proteins modified with N-glycans that are identical to those of embryophytes, suggesting that the P. margaritaceum secretome is more closely related to those of land plants than to those of chlorophytes. The results of this study support the hypothesis that many of the proteins associated with plant cell wall modification, as well as other extracellular processes, evolved prior to the colonization of terrestrial habitats.
