MALDI informed microdissected airway proteomics

ABSTRACT: After performing MALDI metabolomics analysis, we identified airway regions. On a consecutive slide we used laser capture microdissection (LCM) to cut out regions of interest that had an airway specific signal by MALDI. The samples were prepped using the MicroPOTS protocol (PMID: 33491460) and analyzed by LC-MS/MS. The search was performed using MaxQuant and both LFQ and iBAQ quantification were performed.

Human disease states are biomolecularly multifaceted and can span across phenotypic states, therefore it is important to understand diseases on all levels, across cell types, and within and across microanatomical tissue compartments. To obtain an accurate and representative view of the molecular landscape within human lungs, this fragile tissue must be inflated and embedded to maintain spatial fidelity of the location of molecules and minimize molecular degradation for molecular imaging experime  ...[more]
