ABSTRACT: BS3 crosslinking of the human TSEN complex
The tRNA splicing endonuclease (TSEN) complex performs the first step in the maturation of the anticodon stem loop of tRNAs. To date, eukaryotic TSEN complexes have been refractory to structural elucidation and this lack of structural information has hindered our understanding of tRNA recognition and processing. In the manuscript associated with this data, we employed Cryo-Electron Microscopy to solve structures of the human TSEN complex bound to a pre-tRNA in the pre-cleavage state. The structures, solved at near atomic resolution, revealed the overall architecture of the complex and the interfaces involved in tRNA binding. Overall, our data provide critical details into the molecular mechanisms of eukaryotic tRNA splicing as well as an understanding of disease-causing mutations in this protein complex.
