Characterization of the proteome of E. coli and P. aeruginosa from exponential into deep stationary phase

ABSTRACT: The goal of this experiment was to assess changes in the proteome of E. coli MG1655 and P. aeruginosa PAO1 as they go deeper into stationary phase and become more dormant. In this project we grew cultures of these model organisms in glucose-based minimal medium at 37 degree and sampled aliquots to determine their proteome at various growth phases, including 3, 12, 24, and 48 hours after after 1:100 dilution from dense overnight cultures. Biological triplicates of culture aliquots were snap-frozen, and whole-proteome extraction followed by MS/MS detection of peptides was performed.
