Malaria Extracts- natural product libraries grant. positive polarity

ABSTRACT: This was a 1500+ sample run taken in both positive and negative mode. This submission is just for the positive data. These are fungal extracts grown by the Cichewicz Lab, prepared by Thillini P, and run on the McCall Lab Q Exactive Plus for the joint Natural Product libraries building grant. These samples had previously had malaria docking on them. at this point of the study, the results of the docking are kept blind for data analysis.

Project description:Subjects are children from Kilifi District Hospital, Kenya, recruited in 2001 malaria season. total RNA extracted from PaxGene tubes, total RNA and Stratagene Universal reference RNA underwent linear amplification (Message Amp, Ambion). Amplified RNA directly labelled Cy5 and Cy3 and hybridised to 'lymphochip' arrays (print run - lc36n). Scanned on Axon 4000B scanner using GenePix 4.0 software. Clinical sample ID provided in 'Disease State Description' corresponds with Sample ID in Table 1 "Clinical Information for the micro-array samples" in the associated publication. A disease state experiment design type is where the state of some disease such as infection, pathology, syndrome, etc is studied. Keywords: disease_state_design

The RNA content of plasma-derived extracellular vesicles in cerebral malaria

Project description:We sequenced the RNA content of circulating extracellular vesicles obtained from retinopathy- positive (CM-R⁺) and negative cerebral malaria (CM-R⁻) patients and community controls (CC) without Plasmodium falciparum infection. We found that some of the transcripts enriched in circulating extracellular vesicles are highly expressed in the brain compared to other tissues and could have a diagnostic potential in the cerebral malaria context.

Project description:Using genome-wide expression profiles from persons either experimentally challenged with malaria-infected mosquitoes or naturally-infected with Plasmodium falciparum malaria, we present details of the transcriptional changes that occur with infection and that are either commonly shared between subjects with pre-symptomatic and clinically apparent malaria or that distinguish these two groups. Our findings confirm and extend aspects of the earliest responses to malaria infection at the molecular level and which may be informative in elucidating how innate and adaptive immune responses may be modulated in different stages of infection. Transcription profiling of PfSPZ Malaria Vaccine trial subjects in a malaria endemic region

Project description:The Sanaria® PfSPZ Vaccine can confer sterilizing protection against liver stage infection by Plasmodium falciparum (Pf) in malaria naïve individuals. The vaccine consists of aseptically purified irradiated Pf sporozoites. The PfSPZ Vaccine trial in Mali was the first to evaluate the safety and efficacy of this vaccine in a malaria endemic region. Vaccinees received five doses of 2.7 X 105 irradiated sporozoites and the efficacy was measured against naturally occurring Pf Infections in Malian adults during the malaria transmission season.
