A multi-organ proteomic atlas of an extreme aging model part 1 (diaPASEF)

ABSTRACT: 17 organs were harvested from 4 12 week old and 4 84 week old male mice. Proteomics for this set was performed by diaPASEF analysis using an EvoSep 60 speed method.

Muscle fiber type-dependence effect of exercise on genomic networks in aged mice models

Project description:the intervention period was 4 weeks, and treadmill exercises were conducted for all exercise groups. Two sessions per day were administered. A minimum of a one-hour break was allocated between sessions. the 9-week-old C57BL/6 young male mice and 84-week-old mice were purchased from Central Lab. The groups were divided into young control (YC), young exercise (YE), old control (OC), and old exercise (OE) groups. Each group comprised three mice and all mice were subjected to RNA sequencing analysis.

The mouse aging proteomic atlas

Project description:This database provides TMT-labeled proteomic data of aorta (thoracic aorta), brain, heart, kidney, liver, lung, muscle (gastrocnemius muscle), and skin (abdominal skin) of 6, 15, 24, and 30 months old male C57BL/6 mice. In addition to the whole-tissue lysate, low-soluble protein-enriched fraction was also analyzed for heart, kidney, lung, muscle, and skin. Bulk RNA-Seq data are available for brain, heart, kidney, liver, lung, muscle, and skin. The tissues used for transcriptomic analysis and proteomic analysis of whole-tissue lysate and low-soluble protein-enriched fraction were collected from the same mice. All analyses were conducted with 4 biological replicates.

Identification of genes associated with endometrial cell aging.

Project description:Aging of the uterine endometrium is a critical factor that affects reproductive success, but the mechanisms associated with uterine aging are unclear. In this study, we conducted a qualitative examination of age-related changes in endometrial tissues and identified candidate genes as markers for uterine aging. Gene expression patterns were assessed by twice RNA sequencing of uteri tissues from wild type (WT) C57BL/6 mice. First experimental samples were obtained from 5, 8 and 60-75 (aged) week-old WT (n=2, each). Second experimental samples were obtained from 5, 8 and 79 (aged) week-old WT (n=5, each). Gene expression data obtained by RNA-sequencing were validated by real-time PCR. Genes expressing the pro-inflammatory cytokines Il17rb and chemokines Cxcl12 and Cxcl14 showed differential expression between a group, which was composed of 5-week-old WT and 8-week-old WT (young), and aged WT mice. Protein expression levels of the above genes and IL-8, which functions downstream of IL17RB, were analyzed by quantitative immunohistochemistry of healthy human endometrium tissue samples from patients in their 20?s and 40?s (10 cases each). In these secretory phase samples, 3,3?- diaminobenzidine (DAB) staining intensity of IL17RB, CXCL12 and CXCL14 for patients in their 40?s was significantly higher than that for patients in their 20?s, as detected by a Mann Whitney U test. These results suggest that these genes are candidate markers for endometrial aging and for prediction of age-related infertility, although confirmation of these findings is needed in larger studies involving fertile and infertile women.

A single-cell atlas of the aging murine ovary

Project description:Ovarian aging leads to diminished fertility, dysregulated endocrine signaling, and increased chronic disease burden. These effects begin to emerge long before follicular exhaustion. Around 35 years old, women experience a sharp decline in fertility, corresponding to declines in oocyte quality. However, the field lacks a cellular map of the transcriptomic changes in the aging ovary to identify drivers of ovarian decline. To fill this gap, we performed single-cell RNA sequencing on ovarian tissue from young (3-month-old) and reproductively aged (9-month-old) mice. Our analysis revealed a doubling of immune cells in the aged ovary, with T and B lymphocyte proportions increasing most. We also discovered an age-related upregulation of alternative macrophage and downregulation of collagenase pathways in stromal fibroblasts. Overall, follicular cells (especially granulosa and theca) display stress response, immunogenic, and fibrotic signaling pathway inductions with aging. These changes are more exaggerated in the atretic granulosa cells but are also observed in healthy antral and preantral granulosa cells. Moreover, we did not observe age-related changes in markers of cellular senescence in any cellular population with advancing age, despite specific immune cells expressing senescence-related genes across both timepoints. This report raises several new hypotheses that could be pursued to elucidate mechanisms responsible for ovarian aging phenotypes.
