Next Generation Sequencing Analysis of colon transcriptomes in BF638R bsh- and bF638R bsh+ colonized Cdx2Apcf/w mice.

Project description:Bacteroides, that expresses bile salt hydrolase (BSH), is considered as a potential drug target for metabolic diseases. In the present study, BSH-expressing Bacteroides was found to be enriched in two different CRC mouse models and colonization of nonenterotoxigenic Bacteroides species, such as B. fragilis 9343 or B. vulgatus , enhanced CRC growth. Expression of the 9343 bsh gene in B. fragilis 638R, a bsh - lacking strain, enabled more bile acids to escape into the colon and accelerated CRC progression . The presence of BSH activated WNT signaling and upregulated CCL28 expression dependent on b-catenin in colon tumors. The activated b-catenin/CCL28 axis elevated intra-tumororal immunosuppressive CD25+ FOXP3+ T reg cells. Anti-CCL28 antibody reversed microbial BSH amplificationinduced immunosuppression. Inhibition of BSH reduced CRC progression, coincident with suppression of WNT signaling and CCL28 expression. These findings provide a novel insight into the pro-carcinogenetic role of NTBF in CRC and characterize BSH as a potential target for CRC treatment.

Title: Intrinsically disordered RNA-binding motifs cooperate to catalyze RNA folding and drive phase separation Abstract: RNA-binding proteins are essential for gene regulation and the spatial organization of cells. Here, we report that the yeast ribosome biogenesis factor Loc1p is an intrinsically disordered RNA-binding protein with eight repeating positively charged, unstructured nucleic acid binding (PUN) motifs. While a single of these previously undefined motifs stabilizes folded RNAs, multiple copies strongly cooperate to catalyze RNA folding. In the presence of RNA, these multivalent PUN motifs drive phase separation. Proteome-wide searches in pro- and eukaryotes for proteins with similar arrays of PUN motifs reveal a strong enrichment in RNA-mediated processes and DNA remodeling. Thus, PUN motifs are potentially involved in a large variety of RNA- and DNA-related processes by concentrating them in membrane-less organelles. The general function and wide distribution of PUN motifs across species suggests that in an ancient “RNA world” PUN-like motifs may have supported the correct folding of early ribozymes.

Hepatic Farnesoid X-Receptor Isoforms α2 and α4 Differentially Modulate Bile Salt and Lipoprotein Metabolism in Mice

Project description:The nuclear receptor FXR acts as an intracellular bile salt sensor that regulates synthesis and transport of bile salts within their enterohepatic circulation. In addition, FXR is involved in control of a variety of crucial metabolic pathways. Four FXR splice variants are known, i.e. FXRα1-4. Although these isoforms show differences in spatial and temporal expression patterns as well as in transcriptional activity, the physiological relevance hereof has remained elusive. We have evaluated specific roles of hepatic FXRα2 and FXRα4 by stably expressing these isoforms using liver-specific self-complementary adeno-associated viral vectors in total body FXR knock-out mice. The hepatic gene expression profile of the FXR knock-out mice was largely normalized by both isoforms. Yet, differential effects were also apparent; FXRα2 was more effective in reducing elevated HDL levels and transrepressed hepatic expression of Cyp8B1, the regulator of cholate synthesis. The latter coincided with a switch in hydrophobicity of the bile salt pool. Furthermore, FXRα2-transduction caused an increased neutral sterol excretion compared to FXRα4 without affecting intestinal cholesterol absorption. Our data show, for the first time, that hepatic FXRα2 and FXRα4 differentially modulate bile salt and lipoprotein metabolism in mice.
