Multi-Omic Evidence of Brain Region Specific APOE Genotype Perturbations in Alzheimer's Disease


ABSTRACT: Simultaneous extraction of proteins and lipids from human frontal cortex and cerebellum brain tissue was performed using the MPLEx procedure for both control and Alzheimer's disease patients. The isolated proteins were then reduced, alkylated, tryptically digested and evaluated with LC-IMS-MS. The proteomics data was matched to an AMT tag database created from pooled and fractionated samples for peptide identification. The extracted lipids were assessed with both LC-MS/MS and LC-IMS-MS to study isomer differences. The lipids were identified using LIQUID software.

Alzheimer's disease (AD) is a neurodegenerative disease with a complex etiology influenced by confounding factors such as genetic polymorphisms, age, sex, and race.
