Thiol metabolic heterogeneity in immune cells causes distinct molecular mechanisms of toxicity upon chronic lead exposure

Project description:Lead pollution is global, billions of people are chronically exposed to lead. Biological thiols are nature sequesters of lead. Human immune system is highly vulnerable to lead, unfortunately the underlying mechanism remains unknown. Using mass cytometry and mass spectrometry, we systematically identified lead targets in immune cells from mice model of chronic lead exposure. We found CD4+ T and neutrophil are most vulnerable to lead, respectively due to low thiol and high glutathione metabolism. Specifically, in CD4+ T, lack of thiol renders severe damage of ribosome by lead, which we successfully rescued using thiol drugs. In neutrophil, robust glutathione metabolism accumulates lead, which can be passed on to proteins of higher binding affinity. Among these proteins, our evidences suggest lead suppresses immune activation through PKC-like, PE/DAG-binding domain on RAF1 and PKCs. Overall, it clarified the mechanisms of toxicity in immune cells upon chronic lead exposure, which provides valuable information for medical care and public health.

Copper toxicity response influences mesotrophic Synechococcus community structure

Project description:Picocyanobacteria from the genus Synechococcus are ubiquitous in ocean waters. Their phylogenetic and genomic diversity suggests ecological niche differentiation, but the selective forces influencing this are not well defined. Marine picocyanobacteria are sensitive to Cu toxicity, so adaptations to this stress could represent a selective force within, and between, “species” also known as clades. We compared Cu stress responses in cultures and natural populations of marine Synechococcus from two co-occurring major mesotrophic clades (I and IV). Using custom microarrays and proteomics to characterize expression responses to Cu in the lab and field, we found evidence for a general stress regulon in marine Synechococcus. However, the two clades also exhibited distinct responses to copper. The Clade I representative induced expression of genomic island genes in cultures and Southern California Bight populations, while the Clade IV representative downregulated Fe-limitation proteins. Copper incubation experiments suggest that Clade IV populations may harbor stress-tolerant subgroups, and thus fitness tradeoffs may govern Cu-tolerant strain distributions. This work demonstrates that Synechococcus has distinct adaptive strategies to deal with Cu toxicity at both the clade and subclade level, implying that metal toxicity and stress response adaptations represent an important selective force for influencing diversity within marine Synechococcus populations.

Model of heart failure induced by mild thiol-blocking with cystamine

Project description:Depletion of cardiac ATP content is a characteristic feature of heart failure in patients and experimental animal models. To analyze the impact of insufficient ATP supply on heart function we inhibited cellular respiration by disulfide poisoning with the mild thiol-blocking agent, cystamine. We chose 4 month-old apolipoprotein E (apoE)-deficient mice, which are highly vulnerable to increased oxygen and ATP demands. After 4 weeks of cystamine treatment (300 mg/kg in drinking water), echocardiography and histology analyses demonstrated that apoE-deficient mice had developed heart failure with cardiac dilation. The microarray gene expression study of heart tissue from cystamine-treated apoE-deficient mice relative to untreated mice confirmed the development of heart failure showing up-regulation heart failure-specific genes by mild thiol-blocking with cystamine.
